Objective:To study the effects of long-term dysbacteriosis and the recovery of the flora to observe the changes of microbial flora and the influence of the immune system,and to explain the relationship between the development of the intestinal microflora and the host immune system.Bacterial flora to improve the immune function to lay the foundation,also for the clinical long-term immune dysfunction patients with immune function recovery,to provide new ideas.Methods:Choose 60 male Balb/c male mice,and divid into two groups,the antibiotic intervention group and the control group.The mice in the antibiotic intervention group were treated with 200 mg / ml / kg ceftriaxone sodium twice a day between 6-8 hours.After 90 days of continuous administration,the antibiotics were stopped and observed for 60 days,30 days,60 Day,90 days,120 days,150 days.The samples were collected for intestinal flora,immunohistochemistry,immune cells,and immunological detection.PCR-DGGE method was used to analyze the intestinal microflora of mice,and the different bands were cut,purified,sequenced and compared with bacterial species.The levels of serum Ig G,IL-4 and IL-17 in mice were detected by ELISA.Thelevels of SCF,GMCSF and IL-7 in thymus were detected.The levels of s Ig A in small intestine were detected.The contents of T cells and B cells in spleen were detected by flow cytometry.Results:1.Effects of long-term administration of ceftriaxone sodium on body weight of weaning miceCompared with the normal control group,the weight of the mice in the antibiotic-feeding group was increased at 30 days(p <0.05).The weight of the mice in the two groups increased with the passage of time.In the normal control group,after stopping the administration of antibiotics,the weight of the antibiotic group returned to normal.2.Effects of long-term administration of ceftriaxone sodium on spleen index and thymus index in weaning miceCompared with normal mice,the spleen index of the weaning mice was not changed(p> 0.05).The spleen index did not change on the 30 th day,60 days after stopping the ntibiotic intervention(p> 0.05)..3.Effects of long-term administration of ceftriaxone sodium on T and B cells in spleen of weaning miceCompared with the normal group,the content of T cells in the spleen of the mice was not changed(p> 0.05).The content of B cells in the spleen was decreased on the60 th day and the 90 th day after ceftriaxone sodium administration(p <0.05).On the30 th day and the 60 th day after ceftriaxone sodium was stopped,the B cells in the spleen increased,And no difference with normal group(p> 0.05).4.Effect of long-term administration of ceftriaxone sodium on serum immunological factors in weaning miceCompared with the control group,the serum Ig G content of the mice was increased on the 30 th day and the 60 th day after the administration of ceftriaxone sodium group and decreased on the 90 th day(p <0.05).After stopping the administration of ceftriaxone sodium(P <0.01).Compared with the control group,thelevels of serum IL-4 in the mice fed with ceftriaxone sodium group were significantly higher than those in the control group(P <0.01)on the 30 th day and the 60 th day(P<0.01)and increased at 90 days(p <0.05).(P <0.05),and the serum IL-4 level increased at the 30 th day and 60 th day after ceftanoic cementation of cortisone sodium,and the concentration was higher than that of the normal group.Compared with the control group,(P <0.05).The serum IL-17 levels in the weaning mice were higher than those in the normal control group(p <0.05).The serum IL-17 levels were higher than those of the normal control group(p < 0.05),and the serum IL-17 level was significantly increased on the 60 th day(p <0.01).5.Effect of long-term administration of ceftriaxone sodium on thymus immunization in weaning miceCompared with the control group,the content of SCF in the thymus of the decoction mice was lower than that of the control group(p <0.05).The content of SCF was slightly on the 30 th day after stopping ceftriaxone(P <0.05).The content of GMCSF in the thymus of the decoction mice was significantly lower than that in the control group(p <0.05).After stopping the administration of ceftriaxone(P <0.05).The content of GMSCF was significantly lower than that of the control group(p <0.05),but the content of GMSCF was slightly higher than that of the normal control group(p<0.05)),The content of IL-7 increased slightly after stopping cefaclor sodium,but the amplitude was still lower than that of normal control group(p <0.05).6.Effects of long-term administration of ceftriaxone sodium on small intestinal mucus s Ig A in weaning miceCompared with the control group,the content of s Ig A in the intestinal mucus of the mice fed with ceftriaxone sodium was lower than that in the control group(p<0.05).The content of SCF was slightly on the 30 th day after stopping ceftriaxone(P<0.05),and the content of SCF increased slightly after 60 days after stopping ceftriaxone,but the amplitude was still lower than that of normal control group(p<0.05).7.Effects of long-term administration of ceftriaxone sodium on intestinalflora in weaning miceCompared with the normal control group,the number of DGGE electrophoresis bands in the intestinal flora of the weaning mice was significantly decreased(P <0.05)during the administration of ceftriaxone sodium.On the 30 th day after ceftanoic cementation of ceftriaxone sodium,the number of intestinal microflora in the weanling mice was significantly increased.At the 60 th day,the number of DGGE electrophoresis bands recovered and close to the normal control group The level of DGGE electrophoresis bands in milk mice.Conclusion:1.Long-term infusion of ceftriaxone sodium can cause weaning mice intestinal flora imbalance,in the cessation of ceftriaxone2.Long-term flora imbalance can lead to weight loss in weaning mice,spleen index,thymus index did not change,Spleen T cell content has little effect,can lead to decreased B cell content in the spleen.3.Long-term flora imbalance can lead to decreased serum Ig G levels in weaning mice,decreased serum IL-4 levels,and blood Serum IL-17 content increased;bacteria natural recovery,weaning mice serum Ig G content decreased,serum IL-4content increased and higher than normal,serum IL-17 content decreased first and then increased.4.Long-term flora imbalance can lead to decreased thymus SCF content in weaning mice,thymus GMCSF content decreased,Thymus IL-7 content decreased;bacteria natural recovery,weaning mice thymus SCF content content increased slightly high,but the amplitude is still lower than the normal control group,thymus GMCSF content did not change,thymus IL-7 content slightly increased,but the rate is still lower than the normal control group.
