Probiotics play an important role in improving diarrhea,balancing and improving the immunity and regulating intestinal flora.Intestinal tract provides probitics with suvival environment.Symbiotic bacteria in the intestine is accounted more than 90%,it is the main part of the intestinal microbial homeostasis.So,the interaction between the detection about mixed probiotics is a hotspot.Recent studies have shown that although the mechanism of action between many probiotics has been found but the results of these studies have been very different due to the different models.In summary,To explore the interaction about mixed probiotics detection methods and the establishment of a reliable simulation of the intestinal system has become the focus of the field of probiotics research.The research and main results are as follows:The qPCR assay of the probiotic mixed system: Lactobacillus,plantarum,Lactobacillus,salivarius,Lactobacillus,casei Lactobacillus,fermentum,Lactobacillus delbrueckii subsp were mixed and texted.The results showed that the viable count of each probiotic was almost about 107 CFU/mL.In that Lactobacillus fermentum was 4.1×107 CFU/mL,Lactobacillus salivarius was2.2×107 CFU/m L,lactobacillus plantarum was 2.3×107 CFU/mL,lactobacillus casei was 4.1×107 CFU/mL,Lactobacillus delbrueckii was 2×107 CFU/mL.The results showed that the number of viable bacteria was higher than that of Lactobacillus fermentum and Lactobacillus casei.Indicating that in this mixture system of seven probiotics Lactobacillus fermentum and Lactobacillus casei has a competitive advantage.In this study,we compared the method of fluorescent quantitation and qPCR.the method of qPCR detection is superior to fluorescent quantitation in detection of specificity and accuracy.Probiotics Interaction assay:The results showed that lactobacillus casei mixed with Lactobacillus fermentum was enhanced compared with Lactobacillus plantarum and Lactobacillus fermentum.Simulation of intestinal system construction and application: the existingsimulated intestinal system according to functional classification can be divided into three categories: human intestinal chip,industrial in vitro digestion model,experimental intestinal microbial simulation system.Among them,the experimental intestinal microecological system is more conducive to laboratory research,our simulated intestinal system on the basis of its improvement.simplifying the experimental structure,joined the in simulated intestinal culture system.The advantage of our simulated bowel system is that under the same conditions,more intestinal microbes are obtained and the experimental procedure is simple.The simulated intestinal system constructed in this study has four parts:a small fermentation tank,a culture medium and a flow system,a waste liquid outflow system and a power system.There are 43 kinds of intestinal microbes obtained by simulating the intestinal system,belonging to the genus Lactobacillus,Enterococcus,Escherichia coli,Salmonella,Morganella.We then used the intestinal simulation system to study the types and abundance of intestinal microbes in the human body.The work was done as follows: We treated the subjects in two ways,allowing subjects to eat high-fat foods within a week,Intestinal microbes,and then let the subjects within a week of vegetarian food to detect their intestinal micro-organisms,the results are as follows,a week after eating a lot of high-fat food,the culture of intestinal microbes are: Enterococcus,Escherichia coli,milk Bacillus,Salmonella,Morganella.And a large number of vegetarian food after a week,the culture of intestinal microbes are: Lactobacillus,Enterococcus,Escherichia coli.The following conclusions: the short-term changes in the diet,intestinal microbial species remained unchanged,high-fat diet can lead to increased intestinal bacteria.
