Preliminary Study Of Expression And Clinical Significance Of TNF-α And NF-κB P65in Patients With Gastric Cancer In Hehuang Valley

Objective:To detect the expression of TNF-α in serum of patients with gastric cancer,patients with gastritis and healthy people, and NF-κB p65in gastric cancer tissues andnormal tissues adjacent to carcinoma. To study the molecular mechanism of TNF-α andNF-κB p65in the occurrence and development of gastric cancer, and to discuss theirclinical significance.Methods:The expression of TNF-α in serum of twenty healthy subjects, twenty patientswith gastritis and twenty patients with gastric cancer were detected by Enzyme LinkedImmunosorbent Assay. The expression of protein NF-κB p65in gastric cancer tissues andcorresponding normal tissues adjacent to carcinoma of twenty patients with gastric canceras above mentioned were detected by Western Blot. The differential expression ofTNF-αand NF-κB p65were compared and both of correlation and relationship betweenthe occurrence and development of gastric cancer were analyzed.Results:There was a significant difference in the expression of TNF-α in serum of patientswith gastric cancer(twenty cases), patients with gastritis(twenty cases) and healthypeople(twenty cases)(P<0.001), and there is association between gastric cancer stages andmetastasis lymph node(P<0.05). There was a significant difference in the expression ofprotein NF-κB p65in gastric cancer tissues and tissues adjacent to carcinoma(P<0.001),and there is association between gastric cancer stages and metastasis lymph node (P<0.05).Expression of TNF-α and NF-κB p65were positively correlated in gastric patients（r=0.745，P<0.01）.Conclusion:TNF-αand NF-κB p65may be molecular markers for diagnosis of gastriccancer, there is association with clinical stage and metastasis lymph node in gastric cancer.Expression of TNF-α and NF-κB p65may play an important synergy in the occurrenceand development of gastric cancer in Hehuang Valley.
