| Cycle in order to study the expression of cytokines and its relationship with apoptosis and anti-apoptotic genes different mechanisms of luteal cells of the corpus luteum in Yanbian cattle, cattle at different stages of this study was to use an ordinary optical microscope apoptosis corpus luteum corpus luteum morphological characteristics; and the use of SDS-PAGE, Western Blot, PCR amplification and DNA electrophoresis test methods to study different periods luteal cells antiapoptotic genes (Bcl-2, Bax and cFLIP) expression, the following results:1. in the ultrastructural observation, CL apoptotic cell volume becomes smaller, and has an irregular shape, the cytoplasm and condensation gathered together, mainly in the internal nuclear membrane, followed by debris after splitting nuclei are distributed unevenly in the cytoplasm, and finally the formation of a membrane surrounded by apoptotic bodies.2. the anti-apoptotic gene cFLIP, the expression of Bcl-2 in the early and mid-luteal high and low expression of the gene and in the late luteal phase and the difference was not significant.3. The experiment analyzed by RT-PCR technique to see the expression of estrus cycle corpus luteum in Yanbian cattle cFLIP mRNA’s. The results show, cFLIP mRNA expression higher in the early, mid, late and white body. Matsumoto reported as an anti-apoptotic factor inhibiting cFLIP porcine follicular atresia, which is similar to the results of this experiment. From these results, we can guess cFLIP as survival factors involved in luteal degradation. In addition, expression of Bad, Bax, Bid and pro-apoptotic genes such as Fas and Bcl2, cFLIP and Caspase-3 and other anti-apoptotic genes in Yanbian cattle corpus luteum will lay the theoretical foundation for future research. |
