| In order to explore the relationship between steroid secretion regulatory factor LRH-1 and progesterone level in luteal tissues at different stages and expression levels of apoptosis-related factors at four different stages of Yanbian cattle,fluorescence quantitative PCR was used in this experiment.Western Blot was used to detect the expression changes of steroid gene regulatory factor LRH-1 and steroid synthesis related genes STAR,CYP11(P450),HSD3B7(3β-HSD)in luteal tissues of Yanbian cattle at different stages.The correlation between steroid secretion-related genes and progesterone levels in luteal tissues at different stages of luteal degeneration was analyzed.The expression of apoptosis-related genes(bcl-2,BAX,CASPASE-3,Cleaved CASPASE-3)in luteal tissues sampled at different periods of the experiment were analyzed using the same technique.To investigate the change trend of apoptosis gene expression level and the relationship between steroid secretion regulator LRH-1and apoptosis of luteal cells during luteal degeneration.Analysis of test results showed that the expression level of steroid secretion-related factor LRH-1 in the process of luteal degeneration showed the same trend as that of progesterone,increasing at first,then decreasing,and reaching the peak in the middle stage of luteal degeneration,with significant changes in each period(P<0.05).The expression levels of steroid synthesis-related genes STAR,CYP11(P450)and HSD3B7(3 β-HSD)during luteal degeneration showed the same trend and significance as LRH-1.When the expression ratio of apoptotic factor bcl-2 and apoptotic factor BAX in luteal cells was equal to 1,the apoptosis and proliferation of cells reached a balance.When the expression ratio of apoptotic factor Bcl-2 and apoptotic factor BAX in luteal cells was greater than 1,cell proliferation was observed,and when the expression ratio was lower than 1,cell apoptosis was observed.The value of Bcl-2/BAX increased first and then decreased,and the changes of each period were significant(P<0.05).The expression levels of pro-apoptotic factor Cas PSAE-3 and Cleaved CASPASE-3were gradually increased in luteal tissues at different periods,and the changes between different periods were significant(P<0.05),and the apoptosis level was gradually increased.It can be concluded that the expression of steroid secretion regulator LRH-1 in luteal tissues of Yanbian cattle during luteal degeneration is related to the changes of steroid hormone progesterone secretion level and cell apoptosis level in luteal tissues of Yanbian cattle during luteal degeneration.In conclusion,the changes of luteal function in Yanbian cattle are closely related to the regulation of genes related to steroid hormone synthesis.This experimental result provides a theoretical basis for the regulation of reproduction of Yanbian cattle,and lays a foundation for further improving the reproductive function of Yanbian cattle. |
