The test is passed orthogonal design of methods,determine the processof ultrasonic extraction in humulus scandens,using ultraviolet spectrophotometer ofmethods,testing the different parts of contents in humulus scandens.Through in vivoand vitro bacteriostatic tests observe effect of the total flavonoids.To providetheoretical and application on the basis in humulus scandens.Experiment I The extraction technology of total flavonoids in humulusscandens.Using ultrasonic auxiliary ethanol extraction total flavonoids in humulusscandens,through orthogonal design of methods determined extraction yield.Exploreroots,stems,leaves,flowers,mixed,seeds from ultrasonic extraction of total flavonoidsin different times.The results showed that: The biggest impact factors is ethanolconcentration(P<0.01),reach the optimal extration conditious: Extracting30min fromultrasonic,the proportion of raw material and solvent is1:40,the ethanol concentrationis40%,receiving total flavonoids contents is3.47%.Experiment II The bacteriostatic comparison of Gram positive bacteria and theGram negative bacteria of total flavonoids in humulus scandens.In vitro test: The preparation of flavonoids liquid in humulus scandens(contentsis3.47%);Selecting the Gram positive bacteria(Staphylococcus aureus,Streptococcuspneumoniae,Enterococcus) and the Gram negative bacteria(Escherichiacoli,Salmonella);Compare the best bacteriostasis effect.The results showed that:The best bacteriostasis effect is Gram positivebacteria,especially is Staphylococcus aureus.Experiment III Treatment the mices diarrhea is give rise to Staphylococcusaureus in humulus scandens.In vivo tests: Using of intraperitoneal injection of staphylococcus aureus from setup the animal diarrhea,divide into the mices: Blank group;Model group;Positivecontrol group;Low dose of traditional Chinese medicine(contents is3.47%,0.2ml);Thedose of Chinese medicine in the treatment group(contents is3.47%,0.4ml);High dose of Chinese medicine treatement group (contents is3.47%,0.6ml). To observe themices of clinical symptoms,record the change of autopsy,use the technology ofconventional slice to observe the mices from the change of pathological tissue andsmall intestine mucosa.The results showed that: The best treatment effect is high dose of Chinesemedicine treatement group(P<0.01);The small intestine chorionic repair well, thesame of effect with a positive control group.From the overall study results of the experiment,using the ultrasonic extract highlevels of the total flavonoids in humulus scandens,and the well bacteriostatic effectfrom in vivo and vitro.
