Objective: To observe the histopathology change and HSP70level of Spragu-Dawley rat by using Medical hospital late well-known weaponsexperts Professor Zheng Huaixian’s ZhengShiYiHaoXunXiYao in treatment of acutesoft tissue injury. And to study the pharmacodynamic action and machanisms of theZhengshiyihaoxunxiyao, Providing a theoretical basis for clinical treatment.Methods:(1)72three months SD rats, weight220±20g, laboratory adaptivefeeding7days before the heavy blow method made the module. According to thetable of random number method randomly divided it into two groups (n=36), thecontrol group(ZhengShiYiHaoXunXiYao) and the blank control group.; After24hours giving the control group the treatment of ZhengShiYiHaoXunXiYao,2timesdaily, and the control group use the water fumigation treatment, twice a day. Toobserve the index change of the soft tissue injury area and1th days after, the3th day,the5th day: The experimental group and control group were randomly picked up12rats, after death take skeletal muscle paraffin embedding, sectioning,light-microscopy histomorphology changes. And through the immunohistochemistryand immunoblotting method for determining rat gastrocnemius and the change ofHSP70Results:(1) Injury symptoms index observation shows: Compared the controlgroup with the experiment group. The former group has a much better performancethan the latter one.which shows the ZhengShiYiHaoXunXiYao can obviouslypromote the situation of the damage of local swelling, subcutaneous ecchymosisand subcutaneous bleeding dissipation, and significantly improved the state of mindof the rats after the injury.the2thday, there was no significant difference (P>0.05);the4thday, the experimental group index lower than the control group indexdifference between the two groups was significant (P <0.05); the6thday, theexperimental group were significantly lower than the control group index betweenthe two groups, there is a highly significant difference(P<0.01);(2)Light-microscopy:ZhengShiYiHaoXunXiYao has significantly and quickly reduced the inflammationand edema of the injuried area and significantly reduced the process of the musclefiber repairing compared with the model control group,day2, the experimental groupand the blank group no significant difference (P>0.05);day4, between the two groups have a significant difference (P<0.05);day6, between the two groups washighly significant difference (P<0.01);(3)Immunohistochemistry:ZhengShiYiHaoXunXiYao could significantly inhibit the changing of injury softtissue HSP70,in the point of1th、3th、5th day,it gets an obviously differencecompared with the model group.The level of HSP70of the two groups isdecreased.the2thday, there was no significant difference (P>0.05); the4thday, theexperimental group index lower than the control group index difference between thetwo groups was significant (P <0.05); the6thday, the experimental group weresignificantly lower than the control group.index between the two groups, there is ahighly significant difference(P<0.01);(4)immunoblotting method(Western Blot):Compared with the control group,the experimental group has a high expression ofHSP70in its organizations.at three points,between the experimental group andblank group there were statistically significant (P <0.05). According to westernBlotting scan analysis,HSP70’s trend is increased and then decreased,4thday it is onthe peak,6thday it decreased to some extent, but still higher than the expression ofthe value of the2thday.Conclusion:ZhengShiYiHaoXunXiYao can treat the subcutaneous blood stasis,swelling, pain and dysfunction etc caused by acute soft tissue injury.ZhengShiYiHaoXunXiYao may play a key role by increasing the HSP70expressionof muscle tissue.
