Objectives:This study aim to determine astaxanthin ethers hydrolysis and astaxanthin free accumulation in brine shrimps and explore the optimal growth conditions for local Artemia and expand cultivation. Industrial CO2supercritical extraction process are to be optimized. To verify hypoglycemic biological function and hypolipidemic biological function of ASTA. Providing a reference for the new development approach of ASTA.Methods:1. About feed-free adult brine shrimps were randomly divided two groups of brine shrimps each, respectively as break control and experimental groups. Since feed powder of Adonis dry flower in experimental group, samples were obtained to extract by after0.5,1,2,3,4and5hours each group for C18-HPLC and UV analysis.2. Orthogonal experiment whose factors and levels are salinity(1%,3%,5%), pH(8,8.5,9), fodder(Fish meal, Shrimp meal, both) is built to screen the optimal growth conditions for local Artemia and expand cultivation by it.3. We investigated effect to ASTA extraction rate under extraction pressure, temperature, CO2velocity of flow, and use response surface analysis to determine the optimum conditions.4. Diabetic SD rat models are copied by injecting Streptozotocin(STZ), and detect random blood glucose, glycosylated hemoglobin, Superoxide Dismutase (SOD) and Maleic Dialdehyde (MDA) by blood glucose meter, automatic biochemical analyzer and related kit.5. Using high fat fodder copy to hyperlipidemia SD rat model, and detect Total Cholesterol (TC), Triglycerides (TG), High-density Lipoprotein (HDL), and Low-density Lipoprotein (LDL) by automatic biochemical analyzer and related kit.Results:1. The accumulation of astaxanthin free had been increasing for3hours and reached the maximum at the third hour. At the same time, the accumulation of astaxanthin ethers had been descending for3hours until zero.2. The growth of No.3is best; The growth of No.2,4,6,9is better; The growth of No.1,5,7,8is worst.3. Under this condition:extracting pressure-47.0MPa, extracting temperature-49.2℃, CO2velocity of flow-9.0L/h, extracting time-3.0h, we can obtain maximum ASTA extracting rate which up to0.9601%.4. Compared with diabetic model group, the content of random blood glucose, glycosylated hemoglobin and MDA decreased significantly (P<0.01) and the content of SOD increased significantly (P<0.01) at ASTA group.5. Compared with hyperlipidemia model group, the content of TG, TC and LDL decreased significantly (P<0.01) and the content of HDL increased significantly (P<0.01) at ASTA group. Conclusions:1. After feed powder of Adonis dry flower in brine shrimps, astaxanthin ethers were hydrolyzed to produce astaxanthin free which can be cumulated in vivo.2. The Artemia can grow best while salinity is5%and pH is9and the fodder is blend.3. CO2supercritical extraction has high extracting rate and yield.4. Astaxanthin has good hypoglycemic function via decreasing the content of random blood glucose, glycosylated hemoglobin and MDA, and increasing the content of SOD.5. Astaxanthin has good hypolipidemic function via decreasing the content of random blood glucose, glycosylated hemoglobin and MDA, and increasing the content of SOD.
