Nonalcoholic fatty liver disease(NAFLD)is the most common chronic liver disease in developed countries and developed areas in China,and it is also the characteristic of metabolic syndrome in liver.Epidemiological studies show that perilla oil,rich in alpha linolenic acid(ALA),can interfere with the progression of liver injury and lipid metabolism,but the specific mechanism is still unknown.However,ALA is highly susceptible to oxidation,and therefore,ingestion of ALA readily leads to lipid peroxidation.Astaxanthin,as an effective antioxidant,can prevent and control NAFLD in itself.In recent years,circadian clock regulation of liver metabolism has become a hot spot,and may be a possible mechanism for the prevention and treatment of NAFLD.Objective: To establish a NAFLD animal model by giving male C57 bl / 6 mice fed high-fat feed;Researching on the combined effect of perilla oil and astaxanthin alleviates NAFLD;And to Study the effect of perilla oil,astaxanthin or perilla oil with astaxanthin on the regulation of circadian rhythm to alleviate NAFLDMethods: Male C57bl/6 mice were randomly divided into five groups(20 animals per group),and each group is fed one of the following five diets for 12weeks: the first group fed with control diet,normal control group(CON,n=20);The second,third,fourth and fifth groups were fed with high fat diet,respectively,high fat model group(HFD,n=20);The perilla oil intervention group(PO,n=20);The astaxanthin intervention group(AXT,n=20);Perilla Oil and astaxanthin combination treatment group(PO+AXT,n=20).After 12 weeks of feeding,measured their glucose tolerance,and then continue to feed for a week,20 animals in each group were randomly divided into early and late groups.Serum was collected,liver and other tissues were quickly removed and weighed.Serum total cholesterol(TC),total triglyceride(TG),low density lipoprotein(LDL),fasting plasma glucose(GLU),free fatty acids(FFA),alanine aminotransferase enzyme(ALT)and aspartate aminotransferase(AST)activity were determined using respective diagnostic kits.The fixed liver tissues were embedded in paraffin and sectioned thickness,stained with hematoxylin and eosin.The expression of CLOCK,BMAL1 and other biological clock genes in liver tissue and the expression of the lipid metabolism related genes SREBP1,FASN,ACACA,CD36 and the expression of cholesterol metabolism related genes CYP7A1,HMGCR,ABCG5 were detected by qPCR technique;Results: compared with HFD group and CON group,the liver index and fat index increased significantly and serum TC,TG,LDL,ALT,AST were significantly increased;And the liver morphology changes,lipid accumulation and inflammatory infiltration could be easily observed on HFD liver cells,which means NASH had happened.And the perilla oil,astaxanthin,perilla oil and astaxanthin combined intervention group in the detected index showed the effect is obviously improved.The differences between group PO+AXT group and PO group or group AXT group has no significant.High fat feeding leads to liver lipid and cholesterol metabolic disturbance,however the perilla oil,astaxanthin and perilla oil with astaxanthin were able to significantly improve the expression of hepatic lipid metabolism and cholesterol metabolism related genes,such as SREBP1,FASN,ACACA,CD36,CYP7A1,HMGCR,ABCG5 etc..In addition,the group of PO,AXT and PO+AXT can also be used to alleviate or modify the circadian clock related genes CLOCK and BMAL1,which are caused by high fat diet,as well as the disorder of circadian rhythm of early and late hepatic lipid and cholesterol metabolism genes.Conclusion: The perilla oil,astaxanthin and perilla oil combined with astaxanthin can have good control effect on NAFLD induced by high-fat,and can alleviate liver cell injury and metabolic disorders that caused by high fat diet,but adding astaxanthin to perilla oil,which enhances its antioxidant capacity is not having better effect to NAFLD compared with Perilla oil alone,perilla oil,astaxanthin alone or in combination may alleviate NAFLD by regulating the biological rhythm,affecting the liver lipid and cholesterol metabolism.
