| Studying on the hypoglycemic and hypolipidemic effect of Rosa Roxburghii Tratt tea on mice provided scientific basis for the unique wild resources of Guizhou.And researching the extraction of total flavonoids and its antioxidant activity,which can provide the theory basis for the mechanism.The main results were as follows:Purifying method was extracting with water,and it was divided into four different parts: tea powder part;crude water extract part of tea;75% alcohol soluble part of tea;75% alcohol precipitate part of tea.The results showed that the extraction rate of crude water tea was 31%,mainly containing organic acid and other water-soluble substances;the extraction rate of 75% alcohol soluble part was 25%,mainly containing polyphenols,flavonoids and other alcohol-soluble small molecules;the extraction rate of 75% alcohol precipitate part was 6%,mainly containing polysaccharide,protein and other macromolecules.In this study,metformin was used as the positive control,and four parts of high and low dose groups(100 mg/kg,200 mg/kg)were divided into eight groups.After four weeks of continuous administration,studying on the effects of Rosa Roxburghii tea on the body weight,food intake,water quantity,fasting glucose,liver glycogen and muscle glycogen.The results showed that compared with the model control group,the 75% alcohol soluble part of high dose group could significantly reduce the fasting blood glucose level of diabetic mice(P<0.01),and significantly improved the symptoms of diabetes and weight loss in diabetic mice(P<0.01).Compared with the normal control group,the amount of glycogen can be reduced.Studies had shown that Rosa Roxburghii tea has a certain hypoglycemic effect on experimental mice,and its role may be related to its containing tea polyphenols,tea flavonoids and other alcohol-soluble small molecules.In this study,Xuezhikang was used as the positive control,and four groups of high and low dose groups(2 g/kg,4 g/kg)were divided into eight groups.After four weeks of continuous administration,studying on the effects of Rosa Roxburghii tea on the triglyceride(TG),cholesterol(TC),high density lipoprotein cholesterol(HDL-C).The results showed that compared with the model control group,the high dose of water extract of Rosa Roxburghii Tratt tea could significantly reduce triglyceride and cholesterol in hyperlipidemic mice(P<0.01).Studies had shown that Rosa Roxburghii Tratt tea has some hypolipidemic effects on experimental mice,and its effect may be related to water-soluble macromolecules such as tea polysaccharides and proteins.The extraction process of total flavonoids of Rosa Roxburghii Tratt tea was studied by box-Behnken design response surface experiment.The results indicated that the best brewing conditions were material liquid ratio 1:26 g/mL,ethanol concentration 74%,brewing temperature 75 ℃ and brewing time 120 min.In this condition the total flavonoids of tea reached the highest value 2.19%.The total flavonoids of Rosa Roxburghii Tratt tea had strong antioxidant activity,and its DPPH free radical scavenging rate was 0.012270 mg/mL.The DPPH scavenging ability of total flavonoids of Rosa Roxburghii Tratt tea was between Vc and BHT. |
