Objective:Knee osteoarthritis (KOA) is the most common degenerative joint disease in the elderly. The Chinese medicine has unique advantages in the treatment of KOA. Therefore, we have researched on the mechanism of using traditional Chinese medicine compound preparation named Gubi Decoction to intervene the degenerative cartilage cells of C518rat’s knee joint, which not only enriches and develops biological basis and the connotation of modern science that traditional Chinese medicine treat KOA from the impediment, but also establishes the modern biological research ideas and the methodological platform of KOA treatment from rheumatism.Methods:The first, We investigated the regularity of Chinese medicine treatment for KOA in recent10years. At the same time, we selected some normal and degenerative cartilage tissues of human knee joint and degenerative cartilage cells of human being, and also some normal and degenerative cartilage cells of C518rat’s knee joint, observing the morphological characteristics of cartilage cells by a variety of staining methods. Finally, we used MTT to detect the effect of Gubi Decoction on the proliferation of degenerative cartilage cells, which were from C518rat’s knee joint, then we used COL-Ⅱ、MMP-13to dye the degenerative cartilage cells of C518rat’s knee joint, which were intervened with Gubi Decoction, and inquired into its mechanism.Results:(1) Gubi Decoction accorded with the rule of drug using in Chinese medicine treatment for KOA in recent10years.(2) In the degenerative cartilage cells of human knee joint, we saw the primary cells being long fusiform and polygonal, the caryon deformed and atrophied, the passage cells even more. At the same time, the COL-Ⅰ and COL-Ⅱ immunofluorescence in the degenerative cartilage cells of human knee joint were lower expression, but COL-Ⅹ\bFGF、MMP-13immunofluorescence showed high expression. From the pathological changes of human knee degenerative cartilage tissues, we could see the cartilage became thinner, the surface was eroded, and the deep cells became fibroblast-like and mast-like.(3) From the pathological changes of degenerative cartilage tissues, which were from C518rat’s knee joint, we could see the cells being long fusiform and polygonal. With the degeneration aggravating, the proliferation slowed, and the number of cell reduced, At the same time, the cells aged and died.(4) From the effects of drugs on cell proliferation, there were significant differences between the groups with different concentration at the same time (P<0.01). At the same point of time, the Gubi Decoction group and Osaminethacine group had significant difference with different concentration (P<0.01). In physiological saline groups, there were not significant differences between different concentration (P>0.05).In the expression level of COL-Ⅱ and MMP-13, there were significant differences between the groups (P<0.01).Conclusion:(1) Gubi Decoction accorded with the basic theory of traditional Chinese medicine in the treatment for KOA mostly.(2) The COL-1and COL-Ⅱ would be reduced when articular cartilage cells occured degeneration, the cartilage cells primarily became fibroblast-like and mast-like, accompanied with the release of MMP-13.(3) Gubi Decoction could promote the proliferation of degenerative cartilage cells, which were from C518rat’s knee joint, and the cells depended on the drug concentration.(4) Gubi Decoction could regulate the expression level of COL-Ⅱ、MMP-13in degenerative cartilage cells, which were from C5I8rat’s knee joint, thereby delaying the development of KOA.
