Objective:To observe the influence of Gubi granules on rat’s knee cartilage cells and on superoxide dismutase, nitric oxide, malondialdehyde and interleukin-1β of rat’s serum with knee osteoarthritis. Seeking the action mechanism of Gubi granules on prevention knee osteoarthritis. Providing the help for further study. To preliminarily analysis the treatment of typical KOA cases, we try to further illustrate the effectiveness and safety of Gubi granules and lay a foundation for widely used in clinical.Method:40 SD rats were duplicated by Hulth’s technique and randomly divided into the model group, the controlled group (the Celecoxib capsule group), the low dose group of Gubi granules, the dose group of Gubi granules and the high dose group of Gubi granules(n=8), except for the normal group and the sham-operated group(n=8). After the technique, rats were continuously injected penicillin 8×104 U for a week. One week later, rats were enforced to activity 30 min daily for 8 weeks. After 2 days by operated, all rats were given the solution by filling the stomach. According to the humanity and animal body surface area, the equivalent dose was worked out. The controlled group, the low dose group of Gubi granules, the dose group of Gubi granules and the high dose group of Gubi granules were given the solution by filling the stomach.0.9% physiological saline were given the solution by filling the stomach to the normal group, the model group and the sham-operated group continuously for 8 weeks.8 weeks later, Rats’carotid artery blood was extracted. After centrifuged, blood serum of the rats was taken. According to specifications, the content of SOD, MDA, NO and IL-1β were worked out. The right knee cartilage cells in each rat ankle synovial membrane was taken. After 10% formalin fixed and the HE staining, the pathology professionals will check hem.Results:Gubi granules can improve the knee cartilage injury score of rats, effectively prevent articular cartilage damage, and protect the normal morphology of cartilage and synovium. The result was showed by mensutated the content of SOD、MDA、NO and IL-1β in blood serum. Compared with the model group, the content of SOD in the dose group of Gubi granules and the high dose group of Gubi granules group increased significantly(P<0.01), however the content of MDA and NO in the dose group of Gubi granules decreased significantly(P<0.01). And the content of IL-1β in the low dose group of Gubi granules, the dose group of Gubi granules and the high dose group of Gubi granules decreased (P<0.05). There was not obvious difference between the controlled group and the low dose group of Gubi granules, the dose group of Gubi granules, the high dose group of Gubi granules.Conclusion:The result was showed by measured the content of SOD、MDA、NO in rats’ blood serum. Gubi granules can increase the biological effect of SOD, enhance the ability of scavenging MDA、NO and reduce the IL-1β to prevent articular cartilage damage, also can improve the cartilage cell pathological histology form and prevent the further development. In clinical, Gubi granules has obvious anti-inflammatory analgesic effect, can effectively reduce the pain and improve the activity ability of knee joint, with no obvious adverse reactions and high safety.
