【Objective】The study is to investigate the expressions of PLUNC and TLR-4innasal polyps tissue and normal nasal mucosas and their difference, explore thecorrelation between PLUNC and TLR-4, and analyze their role in nasal polypsetiology and disease progression. Our findings may set up the experimentalfoundation for research on the pathogenesis and provide a new idea for clinicaltherapy of nasal polyps.【Methods】All the collected cases of the study were chosen from the patients whounderwent operation in otolaryngological department of Fenyang Hospital Affiliatedto Shanxi Medical University from March2013to November2013, among which45cases of nasal polyps tissue were collected from the patients’ ethmoidal sinuses whosuffered from CRSwNP and underwent nasal endoscopic surgery and30control casesof normal middle turbinate mucosa tissue from the patients who uderwent nasalseptum deviation diorthosis surgery. The expression of PLUNC and TLR-4weredetected by western blot. All data were analyzed by using SPSS17.0statisticalsoftware.【Results】①PLUNC and TLR-4were expressed in both nasal polyps tissue andcontrols. Relative IOD of PLUNC in nasal polyps tissue and controls were0.054±0.022and0.535±0.266separately. Relative IOD of PLUNC in nasal polyps tissueand controls were0.571±0.144and0.080±0.078separately.②In nasal polyps,compared with controls, the expression of TLR-4increased significantly, butPLUNC was the opposite (p<0.05).③There was negative correlation between PLUNC and TLR-4in nasal polyps(r=-0.724,p<0.05).【Conclusion】①PLUNC’s lower expression in nasal polyps than in controlsindicated that limited expression of PLUNC may be another reason for CRSwNP.②TLR-4’s higher expression in nasal polyps than in controls indicated that bacteriamay play an important role in the formation of nasal polyps. The higher expression ofTLR-4may promote the occurrence and development of CRSwNP.③The negativecorrelation between PLUNC and TLR-4suggested that TLR-4may start the signaltransduction pathways which could produce inflammatory mediators and cytokinesto inhibit the expression of PLUNC directly.
