ObjectiveDetect the expression of TSLP in allergic rhinitis (AR) and nasal polyps, and discuss the role of TSLP in the pathogenesis of AR and nasal polyps.MethodsSpecimens were from patients who underwent Nasal Endoscopic Surgery in department of Otolaryngology of Zhongshan Hospital, Xiamen University, including allergic rhinitis 21 cases, 30 cases of nasal polyps and simple nasal septum deviation inferior turbinate nasal epithelial 23 cases. Specimens were divided to three groups, one group was fixed by 4% paraformalclehyde, conventional dehydration, embedded in paraffin, slicing a row, then used to immunohistochemical staining by Elivision detection method in AR and nasal polyps patients with the TSLP expression, the second one was extracted total RNA, reverse transcription for cDNA, while a housekeeper geneβ-actin gene as the internal standard for real-time quantitative PCR reaction, detected in the samples TSLP gene expression level in a separate specimens, the last one was extracted total protein, Western hybridization blot, housekeeper proteinβ-actin protein as the internal standard,Determination with the purpose of the optical density, andβ-actin optical density ratio for representative of TSLP the relative protein expression levels.Results1. Immunohistochemistry results showed that TSLP in the in patients of AR and nasal polyps with a stronger expression, the expression of control nasal mucosa was weak, positive particles were mainly concentrated in the epithelial cells of the nasal cytoplasm. The patients of AR and nasal polyps with TSLP expression was significantly higher than control nasal mucosa (P<0.01), nasal polyps in patients with polyps and the mucous membrane of inferior turbinate there was no significant difference (p> 0.01), and nasal polyps in patients with allergic rhinitis the expression was no significant difference (p> 0.01).2. Real-time quantitative RT-PCR testing revealed that: The patients of AR and nasal polyps with TSLPmRNA expression was significantly higher than control nasal mucosa (P <0.05), nasal polyps in patients with polyps and the mucous membrane of inferior turbinate there was no significant difference (p> 0.05), and nasal polyps in patients with allergic rhinitis the expression was no significant difference (p> 0.05).3. Western blot analysis show that the patients of AR and nasal polyps with TSLP expression was significantly higher than control nasal mucosa (P<0.01), nasal polyps in patients with polyps and the mucous membrane of inferior turbinate there was no significant difference (p>0.01), and nasal polyps in patients with allergic rhinitis the expression was no significant difference (p> 0.01).Conclusions1. From the histological confirmed the TSLP in the AR, nasal polyps showed high expression, mainly distributed in the AR, nasal polyps mucosa cells.2. Real-time quantitative RT-PCR test results showed that TSLPmRNA in AR, nasal polyps were significantly higher than that of control nasal mucosa.3. Through the western blot test, the protein level confirmed TSLP in AR, nasal polyps were significantly higher than that of control nasal mucosa.4. These results suggest that TSLPmay play an important role in the incidence of the AR and nasal polyps, but the detailed mechanism to be studied further. |
