| BackgroundLung cancer is the main cause of cancer deaths in our country. The development of lung cancer results from complicating endogenous and exogenous factors. about the pathogenesy of lung cancer, a number of research focus on gene theory.Recently, someone prefered the theory of danger mode, because they considered that immunologic system recognize only danger signal but not nonself signal .during the procedure of tumor generation and development, it produces lots of danger signal, and then through propotional receptor such as toll-like receptor, these signal are recognized, and then triggering immune response about variant or self antigen. someone indicated that the signal pathway of toll-like receptors may be take advantage of tumor formation, and reported that the up-regulation expression of toll-like receptors have a close relationship with gastric carcinoma and bowel cancer`s genesis and growth. On the other hand,these novel functions of TLRs in tumor biology suggest a new class of therapeutic targets for cancer therapy, and which make it possible to use immunologic adjuvant for anti-tumor immunotherapy. Toll-like receptors are a family of type I transmembrane receptors, they bound pathogen associated molecular patterns (PAMPS), through myeloiddiffer entiation factor 88 and nuclear factor, induce many signal pathways, then initiate cell activation, up- regulate major histocompatibility complex (MHC) and costimulatory molecules expression, secrete cytokine and so on,at last, they induce inflammation response. At now, many reseaches have been tested that in TLRs family,Toll-like receptor4 and toll-like receptor9 have a intimate relationship with tumor formation, but they whether have the same relationship with lung cancer still unknow, which need us to research. ObjectiveThis study was designed to investigate the protein and mRNA expression of TLR4 and TLR9 in lung cancer and tumor-free lung tissues, and analyse the relationship of TLR4 and TLR9 with various clinicopathological such as tumor's histological type, lymph node metastasis, TNM stage. To explore the relationship between expression of TLR4 and TLR9 and oncogenesis development and clinical characteristic of lung cancer.MethodsIn this study, Immunohistochemical methods were used to investigate the expression of TLR4 and TLR9 in 60 cases with lung cancer and 20 cases with tumor-free lung tissues. At the same time, we collected 24 fresh cases of lung cancer and corresponding tumor-free tissues, to detect the mRNA expression of TLR4 and TLR9 by reverse transcriptase polymerase chain reaction (RT-PCR), and the protein of TLR4 and TLR9 by western blotting in lung cancer and tumor-free lung tissues. All the cases were used for routine histopathological diagnosis.ResultsTLR4 and TLR9 positive stained were a greater or lesser degree detected in all of the 60 cases. Positive expression of TLR4 and TLR9 were found 50% (30/60) and 86.7%(52/60) in lung cancer,significantly higher than 10% (2/20) and 15% (3/20) in 20 cases tumor-free lung tissues, evidently (P<0.05). The expression of TLR4 and TLR9 in lung cancer was no relationship with patient's age, gender, smoking and tumor's histological type, lymph node metastasis and TNM stage (P>0.05). but there were close relationship among TLR4 and tumor grade , TLR4 and TLR9(P<0.05).In 24 fresh cases, using western blotting also proved that the protein expression of TLR4 and TLR9 in lung cancer more than tumor-free lung tissues. Meanwhile, by RT-PCR, we found that there were an evident difference mRNA expression of TLR4 and TLR9 in lung cancer and tumor-free lung tissues. ConclusionsAbnormality great expression of TLR4 and TLR9 in lung cancer and the expression of TLR4 are associated with tumor's differentiation degree, the more malignancy, and the more expression of TLR4. All these may hint that TLR4 and TLR9 play an important role in the development and progression of lung cancer, which may provid a new pathway for reseach pathogenesy of lung cancer. |
