Objective:To explore the efficacy and mechanism of Shizao Tang in the treatment of pulmonary fibrosis (PF) on rats through observing the expressions of Tumor necrosis actor-alpha (TNF-a) and Transforming growth factor beta1(TGF-β1) in lung tissues.Methods:108healthy and clean male Wistar rats were randomly divided into six groups as normal control group,model control group, prednisone group, Shizao Tang low dose group, Shizao Tang middle dose group and high dose group. Rats in normal control group was given normal saline instead, while rats in the rest of the5groups induced pulmonary fibrosis with bleomycin endotracheally.Corresponding drugs were given the next day upon the completion of the model. After14and28days of medication,9rats in each group were sacrificed respectively. The pathology of lung tissue was observed microscopically. The expressions of TNF-a and TGF-β1in lung tissues were examined by Immunohistochemistry method. Used SPSS17.0for data statistics.Results:One rat died the next day after given bleomycin endotracheally in Shizao Tang middle group.And another rat in prednisone group died on the9th day. No significant differences was discovered in mortality rates among each group (P>0.05).Compared with normal control group, lung tissues in model control group showed obvious pulmonary alveolitis and fibrosis, and the expressions of TNF-a and TGF-β1in lung tissues were significant stronger (P<0.01). Compared with model control group,pulmonary fibrosis in each treatment group significantly improved,and the expression of TNF-a and TGF-β1were significant decline(P<0.01); Expressions of TNF-a and TGF-β1in lung tissues of rats in control group were much lower than the other groups (P<0.01). The expression of TNF-a in the lung tissue of rats in each group:On the14th day, it was much lower in Shizao Tang high and middle group than in prednisone group (P<0.05);It was much lower in Shizao Tang high dose and middle dose group than in low dose group (P<0.01); No significant differences were discovered between Shizao Tang low dose group and prednisone group,and no significant differences was found between high dose and middle dose group (P>0.05). On the28th day, It was much lower in Shizao Tang high dose,middle dose and low dose group than in prednisone group (P <0.01),and lower in Shizao Tang high dose than in low dose and middle dose group (P <0.01).There was no signigicant difference between Shizao Tang low dose and middle dose group (P>0.05). The expression of TGF-β1in the lung tissue of rats in each group: On the14th day, it was much lower in Shizao Tang high dose group than the other treatment groups (P<0.01),and it was lower in Shizao Tang middle dose group than in low dose group and prednisone group (P<0.01); No significant difference was discovered between Shizao Tang low dose group and prednisone group,and no significant differences was found between Shizao Tang low dose and middle dose group (P>0.05). On the28th day, it was much lower in Shizao Tang high dose group than the other treatment groups (P<0.01),and lower in Shizao Tang low dose group than in prednisone group (P<0.05). No significant difference was discovered between Shizao Tang low dose and middle dose group,and no significant differences was found between Shizao Tang middle and low dose group and prednisone group (P>0.05).Conclusion:Shizao Tang can significantly relieve the symptoms of rats with PF,inhibit the expressions of TNF-a and TGF-β1in lung tissues, improve the quality of life,which can be inferred, Shizao Tang may delaying the process of PF induced by bleomycin trough inhibiting the expressions of TNF-a and TGF-β1.
