Objective: To evaluate the therapeutic effect and mechanism of Buyanghuanwu decoction(BYHUD) in treating pulmonary interstitial fibrosis (PIF) induced by bleomycin in rats.Method: PIF models of bleomycin-induced pulmonary fibrosis were established in rats. 130 SD rats were randomly divided into five groups, including the normal group, the modeled group, the Metreton group, the group I and group II of BYHUD . Expect the normal group had 10 rats, other groups had 30 rats respectively. Then the rats were treated with gastric infusion of normal saline, metreton or BYHUD respectively after 24 hours. Both group were put to death on the 7th , 14th, 28th day except the normal group ,which only put to death on the 28th day. The pathological changes, hydroxyprolin (HYP) level, transforming growth factor β 1 (TGF-β 1) mRNA expression of the lung tissue were measured. Bronchoalveolar lavage were collects and ELISA was applied for TGF- β 1 quantitation. Results: BYHUD can inhibit abnormal heighten of hydroxyproline(HYP),transforming growth factor 3 1 (TGF- β 1) mRNA expression and protein content. Amelioration the pulmonary alveolitis and fibrosis were shown after treatment by BYHUD.Conclusion: BYHUD has definite effect in treating PIF. The mechanism may be through inhibiting the TGF- β 1mRNA expression and protein, decreasing collagen protein, so as to ameliorate the inflammation and fibrosis.
