Objective To investigate the effects of chronic toxicity of Shizao Decoction and Shizao Decoction mixed with licorice on rat liver function and liver, stomach mucosa, so as to improve Shizao Decoction and "Eighteen anti-" the drug safety coefficient, to explore the compatibility mechanism of "toxic Chinese medicine" and "instead of drugs", and provide the scientific the experimental data for the clinical medication safety, promote the Chinese medicine theory of continuous improvement and development.Methods SPF level 90 SD rats, half male and half female, were randomly divided into 7 groups: normal control group, high-dose groups of Shizao Decoction, medium-dose groups of Shizao Decoction, low-dose groups of Shizao Decoction, high-dose groups of Shizao Decoction mixed with licorice, medium-dose groups of Shizao Decoction mixed with licorice, low-dose groups of Shizao Decoction mixed with licorice, normal control group 12, the rest of the group with 13 rats in each group, normal feeding into the cage. 1 weeks after adaptive feeding, rats with equal volume of unequal concentration of gastric perfusion, according to the weight of each administration, 100 g rats were 2ml, the concentration of the high, medium and low-dose group of Shizao Decoction and Shizao Decoction mixed with licorice were respectively 0.28 g, 0.14 g and 0.07 g /(kg?d). The normal control group were given the same volume of distilled water, irrigation and continuous stomach 6 days a week, 1 days off, a total of 90 days. Ninety-first days of withdrawal, the last 1 times after dosing fasted for 24 hours, each group were randomly selected half of rats after anesthesia,abdominal aortic blood test biochemical index of blood, liver, stomach and gross morphology was observed with the naked eye, the changes of liver, stomach tissue structure were observed under light microscope, the remaining half animal observed after discontinuation of 2 week, repeat the above check. In the course of the experiment the rats were observed and recorded daily appearance signs(such as hair color, skin, etc.), behavior, glandular secretion(eye, ear, nose, genital), excreta(color, characters, etc.) changes in diet, weigh 1 times per day 2-3, and adjusted according to the weight of intragastric volume. The analysis of the experimental data by using SPASS20.0 statistical software, P<0.05 had statistical significance.Results1 Weight The experimental results show that, before administration, administration period, recovery period the rats body weight showed no significant difference(P>0.05). 2 Liver function 2.1 Enzymological indicators(ALT, AST) Compared with the control group, rats were administered period ALT, AST were increased(P<0.05), low-dose groups of Shizao Decoction mixed with licorice increased most significantly(P<0.05), suggesting that a long period of perfusion of Shizao Decoction and t Shizao Decoction mixed with licorice can cause cell acute liver injury in rats, due to increased permeability the cell membrane, intracellular enzyme leakage into the blood, resulting in the increase of serum enzyme activity. The recovery period the rats ALT, AST have a certain decline, indicating recovery period of liver injury has improved the trend, but no statistical significance(P>0.05). Compared with the delivery period, recovery period rats of group ALT, AST were lower, indicating liver function has improved the trend, but no statistical significance(P>0.05). 2.2 Protein metabolism function(TP, ALB) Compared with the control group,TP、ALB of rats in low-dose groups of Shizao Decoction mixed with licorice decreased significantly(P<0.05), the other groups were TP, ALB also decreased, but no statistical significance(P>0.05). The reason may be due to liver damage caused by the decrease of protein synthesis ability. Compared with the delivery period, recovery period, rats of group TP ALB has rebounded, but without statistical significance(P>0.05), of which low-dose groups of Shizao Decoction recovered obviously(P<0.05), indicating recovery of rats in each group have a certain degree of liver injury. 3 Pathological changes of liver Compared with the control group, administration of rat liver parenchyma showed various degrees of degeneration(by cell edema, manifested as increased cell volume, cytoplasm loose, pale red dye particles), necrosis(showed nuclear condensation, fragmentation, dissolution the liver cell cord rupture, etc.), interstitial no significant infiltration of inflammatory cells and hyperplasia of fibrous tissue; recovery of rats in each group were no significant liver cell degeneration, necrosis, injury, regeneration of liver cells(see the phenomenon showed increased cell volume, big nucleus anachromasis, binuclear). 4 The change of stomach Delivery period and the recovery period of each group of gastric mucosa in rats with smooth surface, no cover, the mucosal epithelium was single layered columnar epithelium, no shedding, erosion and ulcer, gastric intrinsic layer arranged closely, rules, glandular cavity without expansion, fundic gland found plenty of deep red wall cells and blue purple principal cells, mucous membrane outside each layer is no hyperemia, edema, connective tissue hyperplasia abnormal changes.Conclusion 1 Long-term using Shizao Decoction and Shizao Decoction mixed with licorice had no significant effects on the body weight of rats. 2 Long-term using Shizao Decoction and Shizao Decoction mixed with licorice can cause cells in rats with acute liver injury, for the performance of ALT, AST increased; at the same time the effect of protein synthesis ability, for the performance of TP, ALB decreased. 3 There is a certain relationship between Long-term using Shizao Decoction and Shizao Decoction mixed with licorice effect on liver function and the morphology of liver tissues of rats with dose, the lower the concentration the more obvious toxicity.4 Liver injury in Shizao Decoction and Shizao Decoction mixed with licorice on rats was caused by reversible, liver function recovery period is two weeks after the withdrawal had improved, the degree of injury of liver tissue was obviously reduced. 5 Shizao Decoction and Shizao Decoction mixed with licorice had no effect on the rat gastric obvious pathological.
