Objective:To observe the expression of TLR4and IL-6in the mouse model of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome. By analyzing the expression of the factors mentioned above to explore the possible mechanism of Mieyou decoction in treating helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome.Methods:1. BABL/c mice were divided into control group, model group, only hot and humid group, only Hp group, Mieyou decoction group, Gastric triad groups. The mice of control group were given normal feeding; The mice of only hot and humid group were placed in environments with high-sugar and high-fat diet feeding; The mice of only Hp group were infected with Hp and given conventional breeding; The model group:using the composite factors (hot and humid environment, high-sugar and high-fat diet, H. pylori infection) to make the mouse model of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome; Mieyou decoction group and Gastric triad groups:respectively using "Mieyou decoction" and Gastric triad drugs to treat the model mice. Treatment methods and time of administration is intragastric administration two weeks.2. Using HE staining methods to observe the pathological changes of gastric mucosa of mice3. Using Elisa to detect the expression of IL-6, TNF-a in serum of Mouse.4. Using immunohistochemical to detect the TLR4protein expression in gastric mucosa.5. Using real time quantitative PCR to detect TLR4mRNA in gastric mucosa.Results:The expressions of TLR4protein and mRNA in gastric tissue of mice, IL-6and TNF-αin serum in the model group, only hot and humid group, only Hp group increased, compared with the control group were statistically significant; Those in the model group also increased, compared with only hot and humid group, only Hp group were statistically significant. The general physical condition of the model mice of taking medicine significantly improved. The expressions of TLR4protein and mRNA in gastric tissue of mice, IL-6and TNF-αin serum in the treatment group compare with the model group were statistically significant, p <0.01; The differences between Mieyou decoction group and Gastric triad group showed no statistical sense, p>0.05.Conclusion:Mieyou decoction is better than Gastric triad in improving the general physical condition of the mice; Mieyou decoction can effectively reduce the inflammation of gastric mucosa in mice of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome, The mechanism possibly by killing Hp, reducing the degree of expression of TLR4, reducing the expressions of IL-6, TNF-α, and while promoting the tissue repair, thus exert its therapeutic effect.
