ObjectiveTo research the correlation and differences in the expression of microRNA among spleen-stomach damp-heat syndrome,liver-gallbladder damp-heat syndrome,syndr ome differentiation andhealthy control group in chronic hepatitis B,and find the dif feren expression profile of MicroRNA between spleen-stomach damp-heat syndrome and liver-gallbladder damp-heat syndrome in chronic hepatitis B.Exploring the ess ence of spleen-stomach damp-heat syndrome and liver-gallbladder damp-heat synd rome from the level of molecular biology.To provide the basis for clinical syndro me differentiation and the objective disease of TCM.MethodsUsing the gene chip technology(Agilent miRNAchip)to detect themicroRNAamo ngspleen-stomach damp-heat syndrome.liver-gallbladder damp-heat syndrome,syndro me differentiation andhealthy control group in chronic hepatitis B which were colle cted from clinical units,to observe the differen expression profile of MicroRNA a mong these four syndrome.Using SPSS20.0 system to analyze the data and measu rement data,using GeneSpring software to carry outdifference analysis and functio nal analysis.Results1.There are 101 microRNAs were screened out in the comparison of spleen stomach damp heat syndrome group and healthy control group which 63 high expression,38 low expression,and nine microRNAs with significant differences among them(P<0.05;FC>2.0 times):6 high expression(P<0.05;FC>2.0 times),3 low expression(P<0.05;FC>2.0 times).2.There are 111 microRNAs were screened out in the comparison of liver-gallbladder damp-heat syndrome group and healthy control group which 79 high expression,32 low expression,and 17 microRNAs with significant differences among them(P<0.05:FC>2.0 times):17 high expression(P<0.05;FC>2.0 times).3.There are 116 microRNAs were screened out in the comparison of syndrome differentiation group andhealthy control group which 90 high expression,26 low expression,and 35 microRNAs with significant differences among them(P<0.05:FC>2.0 times):28high expression(P<0.05;FC>2.0 times),7 low expression(P<0.05;FC>2.0 times).4.Comparison between spleen-stomach damp-heat syndrome group,liver-gallbladder damp-heat syndrome group and syndrome differentiation group.There are 2 identical microRNAs in three groups.The spleen-stomach damp-heat syndrome group and the liver-gallbladder damp-heat syndrome grouphave 2 same microRNAs.The spleen-stomach damp-heat syndrome group and the syndrome differentiation grouphave 3 same microRNAs.The liver-gallbladder damp-heat syndrome group and the syndrome differentiation grouphave 7 same microRNAs.Conclusions1.There are differences expression of microRNAin the plasma between spleen-stomach damp-heat syndrome and liver-gallbladder damp-heat syndrome2.There are two potential differences microRNAs in spleen-stomach damp-heat syndromegroup of chronic hepatitis B:hsa-miR-1273g-3p?hsa-miR-4419b.the regulatory mechanism may be related to the mTOR signal transduction pathway and Protein serine/threonine phosphatase activity.3.There are four potential differences microRNAs in liver-gallbladder damp-heat syndromegroup of chronic hepatitis B:hsa-miR-129-2-3p?hsa-miR-21-5p?hsa-miR-1304-3p?hsa-miR-129-1-3p.the regulatory mechanism may be related to the tumer protein 53 and CGMP/PKG signal transduction pathway.
