Objective:Through animal experiments,explore the effect of Shire Zhongzhu Recipe on spleen and stomach damp-heat mice,and explain its mechanism of action,lay a theoretical foundation for Shire Zhongzhi Recipe to treat spleen and stomach damp-heat syndrome,and provide a scientific basis for clinical application.Methods:The experimental mice were divided into blank group,spleen-stomach damp-heat model group,damp-heat damp-heat model of spleen and stomach,low-dose group of damp-heat zhongzuo prescription,middle-dose group of damp-heat zhongzuo recipe,high-dose group of damp-heat zhongzuo,and positive drug group.After administration treatment,hematoxylin-eosin staining method(HE)was used to observe the pathological damage and improvement of mouse gastric tissue;enzyme-linked immunosorbent assay(Elisa)was used to detect gastrointestinal hormones(gastrin,stomach Motilin)content;real-time fluorescent quantitative PCR(RT-PCR)to detect the expression of key genes in the ERK/AKT signaling pathway;Western blot to detect the expression of p-AKT and p-ERK proteins.Results:Compared with the model group,the gastric mucosal damage and inflammatory reaction in the mice in the treatment group with different concentrations of Shire Zhongzhu Recipe were reduced.Compared with the blank group,the contents of gastrin and motilin in the model group were significantly reduced(***P<0.001,**P<0.01).Compared with the model group,the Shire Zhongzhifang high-dose group(##P<0.01)and the positive drug group(#P<0.05)the content of gastrin and motilin increased significantly,and the effect of the high-dose Shire Zhongzhi Recipe group was better than that of the positive drug group.Compared with the blank group,the expression of ERK and AKT mRNA in the model group was significantly increased(***P<0.001,**P<0.01),and the expression of ERK and AKT mRNA in the middle-dose and high-dose groups of Shire Zhongzhufang significantly decreased(##P<0.01,#P<0.05).Compared with the blank group,the expression of p-ERK and p-AKT protein in the model group was significantly increased(*P<0.05,**P<0.01),and the expression of p-ERK and p-AKT in each dose group of Shire Zhongzhi Recipe significantly decreased(#P<0.05,##P<0.01).Conclusion:Shire Zhongzu Recipe can improve gastric mucosal damage and inflammation in mice with spleen and stomach damp-heat syndrome,inhibit the expression of p-ERK and p-AKT,and promote the secretion of gastrin and motilin,it may be the biological basis for the treatment of damp-heat syndrome of the spleen and stomach by the damp-heat zhongshi decoction. |
