Abstract/Summary:
Oral lichen planus(OLP) is a kind of chronic inflammatorydisease,which could implicate in oral mucosa and skins,with unknownetiology.Clinical manifestations of OLP share some features ofautoimmune diseases.The histology of OLP is characterized by asubepithelial limphocytes,increased numbers of intra-epitheliallymphocytes,and degeneration of basal keratinocytes.It has beenidentified that CD8+T cell mainly infiltrate intra-epithelium andsubepithelium,and a CD4+T cell dense infiltration can be detected in thelamina propria.It is accepted that OLP may be a T-cell-mediatedautoimmune disease.Th-17cell is a novel kind subset of CD4+T cell that,and has been first discovered by Park in2005.The presence of Th17cellsand their signature cytokins,IL17,has been demonstrated in tumormicroenvironment, infectious inflammatory states and autoimmunepathology. Recent studies have suggested that Th17cells may also take apart in the pathogenesis of OLP, and the concentration of IL17in theserum of OLP patients varies.A large body of research has indicated thepolymorphisms of IL17relate with the susceptibility of rheumaticanthritis, inflammatory bowel disease, psoriasis and other autoimmune diseases.However,the relationship between the polymorphism of IL17and the susceptibility of OLP has not been reported yet.Objective:To explore the possible relationship between thepolymorphism of IL17A(rs2275913) and the susceptibility of OLP.Methods:This experiment employed a case control method to explorethe possible relationship.The case group comprised of25OLP patients(6erosive OLP patients and19nonerosive OLP pathients).The controlgroup enrolled18healthy volunteers.The target DNA fragments of themwere aggrandized by PCR,and then were taken to genesequencing.Genotyping and allele detection were performed.Subsequentstatistical analysis followed.Results:1.IL17Ars2275913locus had been detected three genotypes AAhomozygote(16.3%),AG heterozygote(44.2%),GG homozygote (39.5%)in this experiment.2.the frenquencies of the AA homozygote and A allele in IL17Ars2275913of erosive OLP patients were higher than those of the controlgroup and the nonerosive OLP patients.3.the detected distribution of IL17A rs2275913genotypes and alleles inthis experiment had no distinct discrepancies with that of Beijing,Tokyoand Utah,but there was a statistically difference with that of Yoruba. Conclusions:1.IL17A rs2275913locus of Jilin Han population has a single nucleotidepolymorphism,with three genotypes detected.2.The polymorphism of IL17A(rs2275913) and the susceptibility ofnonerosive OLP may not have a relationship statistically,but erosiveOLP does.The susceptibility of erosive OLP related with thepolymorphism of IL17A(rs2275913) strongly, with distinct higherfrequencies of AA homozygote and A allele in erosive OLP group.Aallele of IL17A rs2275913may be seemed as a rish factor of erosiveOLP.3.The polymorphism of IL17A(rs2275913) in Jilin Han population has nostatistically difference with that of Beijing,Tokyo and Utah,but adistinct statistical diversity exists between that of Yoruba and Jilin.
