Objective:To explore the possible relationship between the single nucleotide polymorphisms of T helper cytokine genes and the susceptibility of oral lichen planus.Methods:The polymorphisms of Th1 cytokines—TNF-α, IFN-γ, Th2 cytokines—IL-4,IL-10 and Th1/Th2 regulatory cytokine-IL-18 were detected in 151 OLP cases (including 81 erosive OLP and 70 nonerosive OLP)and 143 healthy controls by using the PCR-SSP and PCR-RFLP techniques and the haplotypes of IL-10 and IL-18 gene SNPs were analysed by PAHSE software respectively.Results:(1) Analyze between the OLP patients and the healthy controls revealed that: the genotype frequency of IL-18-607CC and the allele frequency of IFN-γ+874~*T were significantly higher in the OLP group(P=0.001<0.05, P=0.013<0.05);(2) Analyze between the erosive OLP group and the healthy group showed that: the allele frequencies of TNF-α-308~*A and IFN-γ+874~*T were much higher in the erosive OLP group(P=0.013<0.05, P=0.000<0.05); the genotype frequency of IL-18-137GG were statistically increased in the eOLP patients compared with the controls(P=0.023<0.05);(3) Analyze between the nonerosive OLP patients and the controls revealed that: the allele frequency of IL-4-590 ~*T was much lower in the neOLP group and the IL-10-1082~* A was much higher in the neOLP group(P=0.001<0.05, P=0.013<0.05);(4) The haplotye analyses showed that: the IL-10 protective haplotype -1082A/-819T/-592A was significantly increased in the OLP group(P=0.046<0.05); while the haplotyes of IL-18-137G/-607C,-137G/-607A,-137C/-607A were all statistically different between the OLP patients and the normal controls.Conclusion:Our study suggested that the polymorphisms of representative proinflammatory, antiinflammatory and regulatory cytokines were associated with the susceptibility and development of OLP at some extent.
