Objective: The aim of the study was to analyze the expression of Golli-MBP and stromal interaction molecule1(STIM1) in oral lichen planus(OLP) and their relationship with the pathogenesis of OLP.Methods: Tissue biopsies from 39 patients with OLP(21 atrophic-erosive subjects and 18 reticular OLP subjects) and 16 healthy controls were studied using quantitative real-time PCR(q RT-PCR) and western blot. The m RNA expression and protein levels of Golli-MBP and STIM1 were analysed. The expression levels of Golli-MBP and STIM1 were also investigated in paraffin-embedded biopsy specimens from 38 OLP cases and 20 normal oral mucosa cases by immunohistochemical techniques.Results: In OLP patients, the expression levels of Golli-MBP mRNA and protein were significantly upregulated when compared to that in the control group(P<0.001, P=0.009). The expression levels of Golli-MBP m RNA and protein in the reticular group were significantly higher than that in the atrophic-erosive group(P<0.001, P<0.001). With the same trend, OLP patients showed markedly upregulated levels of STIM1 m RNA expression(P=0.004) and protein expression(P=0.005) when compared with healthy controls. The m RNA and protein expression levels in the reticular group was significantly higher than that in the atrophic-erosive group(P=0.006, P<0.001). Expression levels of Golli-MBP and STIM1 were significantly higher in paraffin-embedded biopsy specimens of OLP patients than in normal controls(P<0.001, P<0.001). Golli-MBP-positive cells were mainly located in the basal, spinous, and granular layers and in the inflammatory cells of the lamina propria,staining was granular in the nucleus. STIM1 staining was cytoplasmic and was observed in the basal, spinous and granular layers as well as in the inflammatory infiltrate in the lamina propria. Both of Golli-MBP and STIM1 expressedsignificantly higher in cells in lamina propria of OLP than in normal controls(P=0.001, P<0.001). But the data showed no statistically significant differences in the epithelial Golli-MBP and STIM1 expression levels between the OLP and control groups(P=0.0571,P=0.071).Conclusions: This study indicates that the overexpression of Golli-MBP and STIM1 may play a role in the pathogenesis of OLP.
