Objective:By using high resolution melting curve analysis(HRM)study of shanxi crowd TLR3 gene polymorphism loci rs5743312 single nucleotide polymorphisms(SNPs)and the risk of oral lechen planus(OLP).Methods:Unified the oral cavity and the general condition questionnaire design,according to the survey,choose between September 2014 and October 2014 in Shanxi Province people's hospital of division of the oral lechen planus of outpatient service conform to the requirements and cases in clinical diagnosis and pathological diagnosis of 83 cases of peripheral blood in patients with oral lechen planus as cases,19 cases of men,women,64 cases,selection in the same period,Shanxi Province people's hospital medical center in dental body health extraction of peripheral blood in 87 cases as control group,whole blood DNA extraction,after all the blood sample-20 ?.Randomly selected 10 samples gradient PCR,filtered through the Tm value.Then randomly selected 10 preliminary experimental samples,sure this site can be separated to use high resolution melting curve analysis genotype.Subsequent PCR for all samples,will result in the Light Scanner96 melting equipment,after melting Light Scanner Software application with Call IT-2.0 system to analyze the curve which classification results are obtained.From the classification results of three kinds of curve 2 samples of each group of randomly selected sequencing,verify the classification accuracy.On two groups of genotype,Hardy-Weinberg genetic balance inspection use SPSS16.0 statistical analysis software on two groups of genotype and allele frequency of chi-square test and related risk analysis.Results: 1.The sequencing results consistent with HRM classification results.2.The OLP cases and healthy controls were accord with Hardy-Weinberg genetic laws of balance.3.Erosion type in patients withoral lechen planus,homozygous CC genotype frequency is lower than the healthy controls,TT homozygous genotype frequency was higher than that of healthy controls,with statistical significance.Allele frequency analysis results show that the T allele in cases of oral lechen planus and erosion type oral lichen frequency in the case group were higher than that of healthy controls,show the TT genotype and allele T of samples,the risk of erosion type of oral lechen planus and significantly higher performance for the CC genotype of the sample.Non erosive oral lechen planus patients compared with healthy controls data of statistical significance.Conclusions: HRM technology is a kind of high sensitivity,specific degree good,high flux method of studying the relation between disease and SNP.TLR3 rs5743312 loci polymorphiisms C/T is related to the type of oral lichen planus in Shanxi crowd,especially more closely relationship with erosion type.It haven't found that TLR3 rs5743312 polymorphisms associated with the occurrence of non erosive oral lichen planus.
