Objective: To observe the effects and mechanism of Qu-feng-ding-tong-fang to treatmigraine using pharmacological experiments. Methods:(1) To research the analgesiceffects of Qu-feng-ding-tong-fang using electrical stimulation in rats tail andformalin-induced pain method in mice.(2) Animal models with migraine caused bynitroglycerin and reserpine were build up, the animals’ behavior respondences to the drugwere observed, the content of NO, CGRP, ET,5-HT et al. were detected and analysed.(3)Observation of the effects Qu-feng-ding-tong-fang on blood flow in rats, cerebral pia matermicrocirculation caused by norepinephrine in mice, isolated rat carotid artery.(4) ADP wasused to induce platelet aggregation and collagen-epinephrine were used to inducethrombus, observing the anti-platelet aggregation and antithrombotic effect ofQu-feng-ding-tong-fang.(5) Observation of the sedation and synergies with sodiumpentobarbital of Qu-feng-ding-tong-fang.(6)To observe the hypoxia effect ofQu-feng-ding-tong-fang using normal pressure hypoxia experiment and the breakageinduced acute cerebral hypoxia experiment. Results:(1) Qu-feng-ding-tong-fang can raisethe pain threshold of rats induced by electrical stimulation. For chronic neuropathic paincaused by formalin in mice, Qu-feng-ding-tong-fang can reduce animals’ licking foot time,improve nociceptive behavioral responses of mice.(2) For animal models with migrainecaused by nitroglycerin, Qu-feng-ding-tong-fang can improve the behavioral symptoms ofanimal models with migraine, reduce the content of NO, CGRP, elevate the content of ET,5-HT, and reduce viscosity of whole blood and piasma. For animal models with migrainecaused by reserpine, Qu-feng-ding-tong-fang can extend clotting time, elevate the contentof5-HT.(3) Qu-feng-ding-tong-fang can increase rats’ blood flow volume, ameliorate meningeal microcirculation caused by NA, inhibit vasoconstriction caused by5-HT andKCl.(4) Qu-feng-ding-tong-fang can inhibit platelet aggregation caused by ADP andthrombosis caused by collagen-epinephrine.(5) Qu-feng-ding-tong-fang can shorten sleeplatency, extend the sleep time of mice.(6) Qu-feng-ding-tong-fang can prolong thesurvival time of mice, and improve the ability of mice’ oxygen resistance. Conclusion:Qu-feng-ding-tong-fang has good analgesic effect, can directly improve the symptoms ofmigraine.The Qu-feng-ding-tong-fang also can obviously improve the behavioralsymptoms of animal models with migraine as well as the vasomotor neurotransmitter suchas NO、CGRP、ET、5-HT, increase cerebral blood flow, improve microcirculation,dilate blood vessels, inhibit platelet aggregation and thrombosis, improve blood rheology.In addition, the Qu-feng-ding-tong-fang also has the role of sedation and hypoxia tolerance.So Qu-feng-ding-tong-fang has good prevention and cure effect to treat migraine.
