In this paper, in order to decide the best LED and CCFL light quality ratios thoseare suitable for the peony callus proliferation, we used bud scales of three varieties ofPeony (‘P. suffruticosa ‘Fengdanbai’、 P. suffruticosa ‘Wulongpengsheng’、 P.suffruticosa ‘Huhong’)as explants to investigate the impact of different light qualityratios on callus proliferation. At the same time, the callus of Peony were used asmaterials to study the effects of different concentration of exogenous H2O2onembryonic callus induction from which we could explore the possible conditions oftransformation from callus to embryonic callus. The results were as follows:1. Taking LED and CCFL in different light quality and ratios as the sources forillumination, we studied their impacts on the the ’Fengdanbai’ callus proliferation. Theresults showed that: with different LED light quality proportion, the highest’Fengdanbai’ weight gain ratio emerged when it was70%R+30%B treatment;100%R treatment was the most suitable one for callus proliferation in the CCFL lightquality. The weight gain ratio were:1.1498, with a significant difference from thecontrol.0.7029higher than control.2. Taking LED and CCFL in different light quality and ratios as the sources forillumination, we studied their impacts on chlorophyll,soluble sugar and solubleprotein in the process of callus proliferation. The results show that: the combination ofred and blue light treatment was much better than a single red or blue light treatmenton the callus.70%R+30%B was favorable to the accumulation of soluble sugar andprotein content in LED light; While80%R+20%B was more conducive to theaccumulation of soluble sugars and protein content in CCFL light.3. Taking LED and CCFL in different light quality and ratios as the sources for illumination, we studied impacts of total phenolic content and PPO, POD activity hadon callus proliferation. The results showed that: in LED, CCFL light quality, peonycallus had the highest total phenolic content in the processing of100%B; the PPOactivity in the processing of100%R was superior to other treatments in both quality;POD activity performed a little differently in LED, CCFL light, its best showed in thecontrol group when it was LED light, compared with100%R treatment in CCFLlight.4. The impacts of exogenous H2O2with various concentration on embryoniccallus induction were researched. The results showed that: the formation anddevelopment of embryogenic callus had something to do with the concentration ofexogenous H2O2. Under the treatment of200μmol·L-1H2O2, part of callus transformsinto embryonic callus, microscope observe showed that non-embryonic andembryonic callus were coexistence. Cells of embryonic callus were smaller thannon-embryonic callus and closely arranged, while nucleuses were larger. when theconcentration of H2O2was200μmol·L-1, POD activity and CAT activity were muchlower than the control, while SOD activity was significantly better than the othertreatments.
