The purpose of this research is to develop a better opening light source for the study and production of plant tissue culture,for this ,the ultra bright Red (main wavelength 640nm)& Blue(main wavelength 640nm)LED was used as light emitting materials ,and the 80C51 based intelligent system was selected to realize the control of light quality ,light intensity and light period of the LED light source, supplying the best light environment for the plant tissue culture and the mass production of plantlets. At the same time,CCFL was used to develop the novel CCFL culture frame. Besides, in this study ,the light distribution of different kinds of light source,expecting to supply an reference and help for the related researchers. The Paeonia suffruticosa'Wu Long Peng Sheng'was used as plant materials to study the effect of different light quality of CCFL light source on the growth of the Paeonia suffruticosa in vitro.And the effect of light quality and light intensity of the novel LED light source on the growth of the Paeonia suffruticosa in vitro was also studied .It is exciting that the novel light source of LED and CCFL got a excellent culture effect ,which was obviously better than the convention fluorescent lamp.So, it means that the novel LED and CCFL light source are better tissue culture light soure ,which will be a good replacement light source of convention fluorescent lamp in the future.1. The ultra bright LEDs and the 80C51 based intelligent was used to develop a light quality,light intensity and light period adjustable LED light source culture frame,to supply an light quality, light intensity and light period adjustable freely novel culture frame; The CCFL was used to develop a light quality adjustable CCFL frame ,and the light quality ratio are separately 100%R,80%R+20%B,70%R+30%B,60%R+40B,100%B,W(whiteCCFL),which were used to study the effect of different light quality on the growth of the plantlet and the energy saving effect.2.The main technique parameters of the novel light source was determined ,it showed that the red LED light intensity can be adjusted between 075μmol m-2 s-1 ,the blue LED light intensity can be adjusted between 0166μmol m-2 s-1 .According to the analysis about the light intensity distribution ,it showed that ,none of the distribution of the light intensity is uniform ,and the light intensity in the central part is much higher ,while the outside of its is much weaker. Besides,in the light environment of LED light source 1μmol·m-2·s-1≈16.96Lux,in the light environment of CCFL light source 1μmol·m-2·s-1≈43.05,and in the light environment of convention fluorescent lamp 1μmol·m-2·s-1≈73.30Lux.3.The novel CCFL light source frame was used to study the different light quality on the growth of Paeonia suffruticosa in vitro ,the results showed that:the treatment of 70%R+30%B got the best results,and it showed that a higher ratio of red light is better for the growth of Paeonia suffruticosa in vitro,the red light is good for the stem elongation.Conclusively, the different light quality of CCFL is better for the growth of Paeonia suffruticosa in vitro than the fluorescent lamp treatment.4.The novel LED light source frame was used to study the various light quality and light intensity on the growth of Paeonia Suffruticosa in vitro ,the results showed that: the effect of LED light source frame was better than the convention fluorescent lamp treatment, and the best light quality ratio was 80%R+20%B,while the best light intensity situation was 50μmol·m-2·s-1. |
