The axillaries buds of P.suffruticosa'Taipinghong'were tested as explants. The effects of exogenous hormone,subculturing times,environmental factor(such as light quality and temperature) on rooting were studied; the change of POD, PPO, IAAO activities and phenol contents during the root development were determined; studies on physiological mechanism ,anatomical and morphological of rooting were investigated by paraffin sections.It was Analysised why root-induction is difficult. In addition, were transplanted,and it was discussed that different proportion of transplanting matrix effeceted the rooting tube seedlings on surviving. the results are as follows:1. The effect of DIF on rooting of peony tube seedlings: the rooting of tube seedlings was the best in the DIF of 6℃,and the root rate of the treatment reached the maximum that was 51.85%,and the root rate of the CK was 31.03%.Thereinto, following increasing of the DIF,the tendency of the index of peony tube seedlings including root rate ,rooting index and rooting vigor increased then decreased.No DIF and the oversize DIF was to the disadvantage of the rooting of peony tube seedlings.2. The effect of different treating time on rooting of peony tube seedlings in LED red light: the tendency of the index of peony tube seedlings including rooting rate,rooting index,the average root length increased then decreased,along with the increase of treating time of the LED red light. Thereinto, the rooting rate of peony tube seedlings which was the best was 84%,and the root rate of the CK was only 52%.The difference of them is significant. The results showed that, it was not obvious that the treatment of short time(6 d) availed rooting of peony tube seedlings in LED red light; the index of the treatment of 9 days in LED red light including rooting rate,rooting index,the average root length was the best;while,the rooting rate and rooting index of peony tube seedlings decreased in the long time of LED red light (12,15 d),and the longer time was,the worse plants growthing was.3. The effect of darkness and low temperature treatment on rooting of peony tube seedlings:the rooting rate of peony tube seedlings was highest in 8℃for 3 days which was 90%.As for as different temperature, the rooting rate and rooting index of peony tube seedlings increased along with the rising of the temperature,and the result was optimum in 8℃. As for as different time, the rooting rate and rooting index of peony tube seedlings increased then decreased along with the rising of days, and the result was optimum in the treatment of 3 days.4.The change of the enzyme activity and phenol contents on rooting of peony tube seedlings: the activity of POD appeared peak in the treatmenet of LED red light and darkness and low temperature in the third day ,then fluctuated,and it increased finally. The tendency of PPO activity increased then decreased, thereinto, the activity of PPO in the treatmenet of the LED red light and the darkness and low temperature was higher then the CK. The tendency of IAAO activity increased then decreased, and the activity of PPO in the treatmenet of the LED red light and the darkness and low temperature was lower then the CK. The tendency of phenol contents decreased then increased,and the phenol contents in the darkness and low temperature higher then the CK.5. The effect of different subculture times on rooting of peony tube seedlings:the rooting rate of peony tube seedlings was 66.67 which was the best in subculture one time. The result showed that the rooting rate of peony tube seedlings in subculture was higher than the CK obviously,and the average root number and average root length were different from the CK significantly too.6. Cytology observation on rooting culture of peony tube seedlings:the result showed that root primordium didn't lurk the peony tube seedlings,and the adventitious root stem from the induced root primordium. the root primordium initiated in the 5th day,then the root primordium differentiated and the adventitious roots started to break through the epidermis from the 9rd to 15th day.The adventitious roots already break through the epidermis in the 25th day.the adventitious roots stem from the callus desquamate and vascular bundle.While,the rooting of the blade and blossom bud mainly were from the callus desquamate.
