Paeonia suffruticosa Andr., with a long cultivation history and wild variety, makes it an attractive ornamental and a very high medicinal value. In order to provide the theoretical foundation and scientific technology for the propagation of'Pengzhou Zi', and for perfecting the tissue culture system in virto of Paeonia suffruticosa Andr., and commercial production in a large scale, in this plant regeneration system study on Paeonia suffruticosa 'Pengzhou Zi'was developed by using axillary bud from 2-3 ages Paeonia suffruticosa 'Pengzhou Zi'as explants, and by means of orthogonal experiment design, completed combination experiment design, and by exercising the analysis of variance and the multiple comparisons, to set the best selection of culture mediums in different cultures stage, including restrain browning culture, inducement and multiplication culture, rooting culture and transplanting stage, and to explore the different influence of different basal culture mediums, growth regulators and concentrations, restrain browning inhibitors, transplanting mediums. The results of this study showed the follows:1. The influence of the explant isolation date, dark culture, AC, VC, AgNO3 to restrain browning phenomenon in the restrain browning experiment showed that, the explant isolation date, dark culture, AC, AgN03 can efficiently control the browning, but VC can not.The restrained browning measure that the axillary buds, which isolated on February, inocalted on the AC medium for 7 days, then shifted to the medium which have no AC for go on the next culture for inducement and multiplication was the best one. In this way, the browning should be efficiently controlled, also avoid the reduce, when AC existed in medium, of the rate of inducement and multiplication.2. The best axillary bud inducement and multiplication medium was MS+6-BA1.0 mg/L+NAA0.1mg/L, in this way, the rate of inducement was 95.37%, and multiplication was 4.34.6-BA and NAA had extremely significant effected on the rate of inducement and multiplication, the more high concentration of 6-BA, the more high rate of inducement and multiplication, but when the concentration of 6-BA get exceeded could make the test-tube seedings vitrifed.3. By using orthogonal design L16 (45), the result of rooting experiment showed that, the basal medium and IBA had extremely significant effected on the rate of rhizogenesis, NAA, the basal medium, and IBA had extremely significant effected on the quality of rhizogenesis, nevertheless, IAA had no significant effect on this.The best rooting medium was 1/2MS+NAA0.4mg/L+IBA2.0mg/L, while the rate of rhizogenesis was 88.89%, it made main root longer and branch root more. Besides, there were toothlike objects around the edge of callus of which some test-tube seedings that would had not differentiate rhizogenesis.4. Up to now rhizogenesis is the biggest difficulty of Paeonia suffruticosa Andr. tissue culture in the world. In this experiment, after 5 harden days, the rooting regeneration plants were shifted to four mixed transplanting mediums, after 20 days the result showed the best transplanting mediums was river sand, vermiculite, wood chips(1:1:1), the rate of survivals was 46.88%.
