The Preliminary Research On Tissue Culture Of Paeonia Suffruticosa

Paeonia suffruticosa is one of the traditional ornamentals .In order to abtain multiplication, Certain factors such as diffevent breeds , diffevent plant growth regulators , diffevent physiological and chemical influences were tested for their effects on Paeonia suffruticosa A's bulbil in the course of germination , multiplication and inducing root by means of Orthogonal experiment .The main compositions of culture medium and their availabile levers were tested . Using tender leafstalk as explant ,the main influences and their availabile levers were tested during callus inducing , multiplication and diffeventiation .at same time the paper discussed the method and the best antioxidant or sorbent inhibiting callus from browening .Analyzing the results are as follows:1) Explants were sterilized with 0.1%Mercurie chloride (HgCl₂.The result showing : To some extently different materials were treaded for different time. The suitable time of tree peony scale bud is better with 5-7min. the time for Petiole bud is short than scale bud.Time is better with about 5min.2) Paeonia suffruticosa A 'Wu Long Peng Sheng' has the highest multiplication among six breeds (P. suffruticosa 'LuoYang Hong' ,P. suffruticosa 'Hu Hong', P. suffruticosa 'Rou Fu Rong' P. suffruticosa 'Lu He Hong' P. suffruticosa 'Wu Long Peng Sheng' P. suffruticosa 'Zhu Sha Lei' )3) Through the scale bud's culture of P. Suffruticosa'WuLong Peng Sheng'.the effects of the diffevent plant growth regulators(NAA , IAA å’Œ 6-BA) and 6-BAsal medium( 1/2MS, MS, WPM å’Œ MS+Ca²⁺(WPM))on scale buds' multiplication were anailyzed .the result showing: the effects of factor s on the multiplication of scale bud , NAA>6-basal medium >6-BA>IAA ; the best shoot prolifcration was abtained with medium MS+CA2+(WPM) supplanted with 6-BA2.0 mg·L^-1 + IAA0.3 mg·L^-1 1 and Ms supplented with Ca²⁺(WPM)+6-BA 1.0 mg·L^-1+IAA0.1 mg·L^-1 The period of succesive transfer culture should be less than 35 days.2) During the rooting , the results of the rate of root, the average number and length of roots showed that: IBA is the effective factor ; the best medium is 1/2MS supplanted with IBA1.0 mg·L^-1+IAA1.0 mg·L^-1 and sugar 20 g·L^-1 for the root induced .basal medium and the sugar's consistency are main influence in root length . the best medium is WPM...
