Effects of different kinds of growth regulators, light conditions, varieties and explants types on the callus induction were studied, the petiole disparted the nearly stem end, the middle and the nearly leave end, leaves, stem as explants, on Paeonia Suffruticosa'Luo Yang Hong','Wu Long Peng Sheng','Hu Hong'plantlet in this experiment. It also studied the effect of sugar concentration,light source and various of plant growth regulators on the anthers and petals of Paeonia Suffruticosa'Luo Yang Hong'for callus induction. Besides, to explore the suitable specials and concentration of hormone on the induction of Paeonia Suffruticosa somatic embryo ,the callus of Paeonia Suffruticosa'Feng Dan Bai'was used to induce somatic embryo. The results were as follows:1. The petiole ,leaves, stem of the Paeonia Suffruticosa'Luo Yang Hong'plantlet were used to induce callus in this experiment ,the best media for the cultivation of near the stem petiole and middle end petiole was 6-BA or 6-BA+2.4-D added media, the highest induction rate was 100%;For the cultivation of middle end petiole was 6-BA(2.0mg/L)and 6-BA(0.5mg/L)+2.4-D(2.0mg/L)added media, the highest induction rate was 100%;For the near leaves end was (0.5mg/L)+2.4-D(2.0mg/L)and 6-BA(0.5mg/L)+NAA(1.0mg/L)added media, the highest induction rate was 100%;And for the cultivation of the leaves was 6-BA(0.5mg/L)+NAA(0.5mg/L) added media, the highest induction rate was 94.4%;For the cultivation of stem was 6-BA(0.5mg/L)+NAA(0.5mg/L)and 6-BA(0.5mg/L)+NAA(2.0mg/L) added media, the highest induction rate was 100%.2. The petiole,leaves, stem of the Paeonia Suffruticosa'Feng Dan Bai'plantlet were used to research different kinds of light quality effect on the induction of callus. The best light quality for the near the stem end of petiole, middle end petiole , near the leave end of petiole were respectively 100% B and R:B= 75:25, R:B= 75:25 and 100% B, 100% B and 100% R. The best light quality for the leaves was R:B= 50:50. The control group proved to have the worst induction rate. 3. The petiole and leaves of Paeonia Suffruticosa'Luo Yang Hong','Wu Long Peng Sheng','Hu Hong'three varity of plantlet were used to induce callus. The results showed that: in point of variety, the induction effect of Paeonia Suffruticosa'hu hong'was the best, next came Paeonia Suffruticosa'Wu Long Peng Sheng', the induction effect of'Luo Yang Hong'was the worst. In point of the types of explants, the petiole near the stem end was the best, next came the petiole near the middle end, the petiole near the leave end proved to be even worse. The induction effect of the leaves was the worst.4. The pollen collected from the flower of flower forcing Paeonia Suffruticosa'Luo Yang Hong'was cultivated at different concentration of sugar and light source,the results indicated that :the best concentration of sugar was3%,6%,but 9% concentration of sugar was not suitable for the pollen cultivation of Paeonia Suffruticosa; In point of cultivation light source, the effect of the weak scattering light was best ,next came the dark condition, the effect of the fluorescent lamp was the worst. The suitable media for the pollen cultivation of Paeonia Suffruticosa'luo yang hong'was MS+PVP+6-BA(1.0mg/L)+2.4-D(2.0mg/l),and the suitable media for the petal cultivation was MS+ PVP +2.4-D(2.0mg/L).5. The callus of Paeonia Suffruticosa'Feng Dan Bai'was used to induce somatic embryo , globular embryo was observed. The highest induction rate was 23.1%. the media was : 1/2MS+PVP +NAA(0.1mg/L)+TDZ(0.1mg/L).
