Liposome encapsulated hemoglobin is a microcapsule small particle that mimics the structure of red blood cells and encapsulates hemoglobin in the phospholipid material.It possesses the function of hemoglobin.Due to the absence of glycoproteins on the outer membrane,the use of LEH is not limited by the blood group.Today,blood products are scarce,whether it is human medicine or animal medicine.There are also many drawbacks of blood transfusion,such as cross-infection of the disease,hemolytic reaction of blood transfusion,etc.LEH is expected to overcome blood disorders in humans and animals.The main contents of this study are divided into four parts:optimizing the preparation of LEH method,exploring the best preservation conditions,LEH in vitro and in vivo safety testing,LEH resuscitation animal models of acute hemorrhagic shock.?1?The effects of each condition on the entrapment efficiency in the preparation of LEH were examined one by one,and the highest enveloping factor was selected as the target.When hemoglobin concentration(CHB=140 g/l),m?lecithin?:m?cholesterol?=2:1,V?chloroform?:V?ether?=3:1,V?oil phase?:V?water Phase?=3:1,rotation rate=100 r/min,water bath temperature=25°C,LEH achieved maximum entrapment efficiency?47.35±2.05?%.?2?The leak rate and color change of LEH at different temperatures in different preservation solutions within 30 days were measured and observed.The results showed that the leakage rate of LEH in the 0.9%NaCl solution was the lowest?18.78%?at 4°C.The LEH particles were still red and capable of carrying oxygen.?3?There was no growth of bacteria in the ordinary broth medium of LEH.There was no coagulation or hemolysis in the blood test with rabbit blood.The LEH was imported into a healthy rabbit and physiological characteristics returned to normal within a short period of time,indicating that it can be safely used in rabbits.However,the increase in blood routine,urine routine,and biochemical indicators indicate that LEH has certain side effects on the body's immune system,liver,and kidneys.?4?In order to investigate the clinical effect of heterologous LEH resuscitation on acute hemorrhagic shock in cats,resuscitation was performed with RBC,LEH,and saline,respectively.We Observe and compare the blood oxygen saturation,blood routine,and biochemical indexes before and after treatment in the 3 groups.The results showed that after3 hours of resuscitation,heterologous LEH can achieve the effect of approximate RBC can modify the oxygen deficit in acute hemorrhagic shock cats and reduce multi-organ damage caused by hypoperfusion,superior to the use of saline.In this study,the preparation,preservation,safety,and functionality of LEH were investigated,which laid a certain foundation for LEH to become a common red blood cell in animals.
