Objective:In order to prepare the artificial red blood cells which were consistent with the requirements of blood substitutes in the aspect of physical characteristics, biological activity and side effects, we evaluated the quality of liposome encapsulated swine hemoglobin which including morphology, entrapment efficiency, oxygen carrying capacity, safety, stability and effective application value.Method:1.The purification and preservation of swine hemoglobin;2.The screening of the optimum prescription and the best preparation conditions of LEH by the method of rotary evaporation, film hydration ultrasonic oscillation and orthogonal test;3.The evaluation of LEH including morphology, entrapment efficiency, oxygen carrying capacity, safety, stability and effective application value by the method of microscopy, spectrophotometer, monitoring the blood oxygen saturation, blood routine test, blood biochemistry, urine routine and the important metabolic organ tissue sections;4.The comparison analysis of effective application value by the emergency treatment of hemorrhagic shock animal.Result:1. The concentration of swine hemoglobin is0.2g/ml after purification; The volume ratio of the oil phase to aqueous phase is3:1; The mass ratio of Hb to the lecithin is10:1; The mass ratio of Cholesterol to lecithin is l:2;The temperature of rotary evaporation is30℃;2. LEH encapsulation efficiency can be stabilized at (41.87±0.46)%; LEH have a certain ability to carry oxygen and can be stored in4℃environment for30days; All the test items of experimental animals can return to normal state in a short time; There is no obvious abnormal in the liver and kidney tissue slice of experimental animals;3. LEH has a certain application value.Conclusion:The entrapment efficiency of LEH could be stabilized by the scheme of preparing artificial red blood cells in this study, and the quality evaluation of LEH was also performed well, indicated that LEH can strive for more time for the further treatment measures in clinical emergency. LEH has a great potential in the study of blood substitutes and is expected to achieve large-scale industrial production of artificial red blood cells...
