Osmanthus cooperi Hemsl., blonging to Oleaceae Genus Osmanthus,is a famous woodyflower speeies and some aspects of research of Osmanthus cooperi is still a blank. In order toprotect it, the geographical distribution, leaf phenotypic variation and genetic diversity ofOsmanthus cooperi have been studied in this paper. The major work and conclusions of were asfollowing:(1) Osmanthus cooperi was subtropical distribution, and the horizontal distribution ofOsmanthus cooperi was mainly in the Yangtze River basin and the south provinces. the locationsof this species were23°-32°N,104°-121°E; vertical distribution of Osmanthus cooperi wasmainly between400to800meters, it could be higher than2100meters in the Xingdou mountainof Hubei Province, but some of place could be lower than200meters, such as inYixing Jiangsuprovince the vertical distribution could lower to65meters. The original place of Osmanthuscooperi were in the Southwest China in Neogene Period to Quaternary period; East China is themodern distribution center of Osmanthus cooperi,and there were two ways for Osmanthuscooperi to distribution.(2) The morphological of leave was discussed by the statistics and numerical taxonomy, andthe result showed that:1) from6leaves phenotypic traits measured values of Osmanthus cooperican concluded that there are some differences in leaf phenotypic traits of osmanthus cooeri, buttheleaf length and leaf width was positively correlated(y=0.1807x+1.5146, R2=0.5222, p<0.001),and the leaf size of Osmanthus cooperi is Jinhua> Nanjing> Huangshan.2) Cluster analysis andprincipal component analysis showed that there were considerable variances in phenotypiccharacteristics of Osmanthus cooperi, but the quantitative variation of leaves were not relativelystable, and the variation between populations and within populations were with great variabilities.Through the contrastion between Osmanthus cooperi and Osmanthus fragrans, found that therewere differences in the characteristics of the veins and flower colors, between Osmanthuscooperi and Osmanthus fragrans, and it could be used as an important reference for thetaxonomy.(3)In the paper, there were12ISSR Primers which were applied in genetic diversity andsystematie relationship of Osmanthus cooperi. The number of amplified fragments was110, andthe average number of each primer is9.17. While the number of polymorphic fragments was98,the percentage of polymorphic fragments was89.10%. Primer UBC815, UBC825, UBC844,UBC853have the highest Pereentage of polymorphic fragments, it’s100%, Primer UBC856hasthe lowest percentage of polymorphic fragments, it’s77.78%. The ISSR cluster results reveal that the relationship of Osmanthus cooperi between Huangshan and Jinhua was relatively close.(4)There were11SSR fluorescent primers which were applied in genetic diversity andsystematie relationship of Osmanthus cooperi. The number of alleles was181, and the averagenumber of each primer is16.45, primer OFP019has the best amplification effect, it has26alleles,primer OFP023has apoor amplification effect, it only has8alleles. The SSR cluster resultsreveal that Osmanthus cooperi and Osmanthus fragrans could be fully distinguish with thethreshold of0.83,32Osmanthus cooperi samples could be preferably distinguished with thethreshold of0.87. There was cross clustering in parts of Osmanthus cooperi in Huangshan andJinhua, it shows that there are some differences in the genetic structure of smanthus cooperi andOsmanthus fragrans,and the relationship was relatively distant, while the relationship ofOsmanthus cooperi between Huangshan and Jinhua was relatively close.
