Hemerocallis fulva has abundant germplasm resources, a widely scattered, a wide range ofvarieties and its valuable in use. In this study, use ISSR markers to study the genetic diversity ofthe genus of Hemerocallis and build fingerprint construction. Experimental results were asfollows:1. This experiment used the technology of ISSR-PCR analyses the phylogenetic relationshipsand genetic diversity between100materials of plants of the genus Hemerocallis include3original species and97cultivars; used ameliorate CTAB to extract the genomic DNA, andan optimal reaction system of ISSR-PCR was established, the system was:25μl systeminclude DNA template1.5μl, primers1.0μl, Taq DNA polymerase0.20μl, dNTP1.5μl,10×PCR buffer2.5μl, ddH2O18.3μl;12primers with better amplification effect were selectedfrom100primers, and determined the best annealing temperature of each primer,12primersamplified144bands which including140polymorphic bands, PPB reaching97.22%, whichmeans that the material used in the experiment has good genetic diversity.2. Then analyze the result of the experiment through the software of PopGene, the resultshowed that: between100parts of test samples efficient allelic number was1.8565, Nei’sgenetic diversity index was0.4557, Shannon’s information index(I) was0.6466, the geneticdistance was between0.0946-0.6523, it shows that the material has a wide coverage of geneand a large interspecific variation; the cluster analysis by NTSYS shows that when geneticdiversity index was0.69, the samples into3classes, first class was No.1-18、No.20-31, andNo.33-50, the second class was No.32, No.51-100; the third class was No.19. Clusteranalysis showed results: H. Little Business, H. Foshou, H. Buffyis Doll, H. DaringDeception, H. Zhujin and H. Rosy Return are belong to H middendorfii Trautv. et Mey.; H.Chaoyang belongs to the hybridization species of H.lilio-asphodelus L.3. The original species H.fulva L and H. lilio-asphodelus L were not into the same class, whichis as same as the research result of Xiong Zhi-yan et. al、Zhu Hua-fang et. al and Li Hai-li et.al, but the result was different from Zhu Yun-hua et. al, this should be related with thedifferent source of experiment material and the different primers.
