Jasminum belongs to the Jasminoideae family of Oleaceae,which is widely welcomed for its beautiful flower shape,aromatic and high ornamental value.At present,there are few comprehensive studies on phenotypic characters and molecular markers of Jasminum,especially on the classification of varieties and genetic diversity.In this study,29 species of Jasminum were evaluated by phenotypic characters combined with ISSR molecular marker technology,in order to provide a basis for the study of species classification and genetic diversity of Jasminum.The main results were as follow:1.The basic statistical analysis of phenotypic showed that:The variation coefficient of phenotypic characters was 23.81%?88.08%,which indicated that there was a great variation in phenotypic characters.The principal component analysis of phenotypic characters of floral and leaf of Jasminum materials showed that:12 floral characters belong to four principal,among which the number of flower petals,bud diameter,100-floral weight and inflorescence type contributed the most to the principal component analysis;9 leaf characters belong to three principal components,among which the leaf length,leaf width,vein number and petiole length contributed the most to the principal component analysis.These phenotypic characters have reference value in classification of Jasminum materials.Through comprehensive analysis of floral and leaf characters,Jasminum NO.13 with the best comprehensive characteristics and the highest ornamental value.2.The results of cluster analysis of phenotypic characters showed that:Jasminum materials could be clustered into three groups at 15 Euclidean distances,and the characters of bud diameter,100-floral weight and corolla length were the main basis of phenotypic clustering.3.The genomic DNA of Jasminum was extracted by improved CTAB method,high purity and quality DNA template was obtained,which met the requirements of ISSR-PCR amplification.Single factor and L16(45)orthogonal design were used to determine the 25?l ISSR-PCR optimum reaction system of Jasnminum:dNTPs 0.10 mmol/L?Tag DNA polymtherase 2.5 U.primers 0.7?mol/L?template DNA 2.5 ng?Mg2+ 1.0 mmol/L;the best reaction system was used to screen 8 ISSR primers with clear amplification,high repeatability and good polymorphism.Of these 8 primers,101 bands were amplified,99 were polymorphic,and polymorphic ratio was 98.16%.4.The results of ISSR marker cluster analysis showed that the genetic similarity coefficient of Jasminum was 0.465-1.000,indicating that 29 materials had abundant genetic diversity.VWhen the genetic similarity coefficient was 0.562,29 materials were divided into three classes.When the genetic similarity coefficient was 0.606,29 materials were divided into 7 sub-classes.5.The results of ISSR clustering analysis and phenotypic clustering analysis showed consistency among most materials:In the ISSR marker cluster analysis,Jasminum No.1,No.4,No.5,No.24 and No.30 were clustered in the same class,which was consistent with the results of phenotypic cluster,indicating that clustering analysis based on ISSR molecular markers was feasible for genetic diversity analysis of Jasminum materials.And there were differences among individual materials only.There were differences between ISSR marker clustering analysis results and phenotypic clustering analysis results of Jasminum No.13 and No.36.The reason may be that those two materials were both Jasminum sambac with similar genetic background,but the difference of provenance environment leads to the large difference of phenotypic characters between them.
