Objective:To observe the effect of compound psoralea corylifolia liniment and sanbai granule on melanocytes and tyrosinase of guinea pig.To explore the mechanism of therapy with compound psoralea corylifolia liniment and sanbai granule on vitiligo.Methods:1Animal experiments:Selected36cases of guinea pigs were randomly divided into A group(n=30) and B group(n=6).The sanbai granule were for oral administration on A group guinea pig. The normal saline were for oral administration on B group guinea pig. A group was randomly assigned to five groups. There were six in every group. The different five back area of guinea pig skin were selected in anyone A group.The2%、4%、6%、8%compound psoralea corylifolia liniment and normal saline were for external use on every group. Before treatment and after28days, we detect the melanin index of the different back area of guinea pig skin through the CR-300color difference meter. The results were compared. We proceed the pathological section of the different area of guinea pig back skin. The cell count of containing melanin granules and melanin content index of were microscopically detect through Schmorl staining. By the the Imokawa staining, melanocyte proliferation of different concentrations of drugs region were microscopically detect. Tyrosinase in the plasma content that blood samples were collected by abdominal aortic of A group and B group were detected.2In clinical observation selected60cases of vitiligo patients who Suffering from head and neck were randomly divided into experimental group (n=30) and control group (n=30).Experimental group:oral sanbai granule, external concentration of6%compound psoralen liniment and narrow-band ultraviolet irradiation. Control group:the narrow-band ultraviolet irradiation. Tyrosinase levels were measured and analyzed before and after treatment. Clinical efficacy was determined. The date was statistically analyzed by SPSS19.0package.Results:The measured melanin index of2%,4%,6%,8%of concentration of compound psoralen liniment application group after28days were compared with those measured before treatment in A group. The difference was statistically significant (p<0.05).The measured melanin index of6%,8%of concentration of compound psoralen liniment application group were compared with2%,4%of one. The difference was statistically significant (p<0.05). The measured melanin index of6%of concentration of compound psoralen liniment application group were compared with8%of one. The difference was not statistically significant (p>0.05).The melanocyte counts of2%,4%,6%,8%of concentration of compound psoralen liniment application group concentration were compared with the saline control group. The difference was statistically significant (p<0.05).The melanocyte counts of6%,8%of concentration of compound psoralen liniment application group were compared with2%,4%of one. The difference was statistically significant(p<0.05).The melanocyte counts of6%of concentration of compound psoralen liniment application group were compared with8%of one. The difference was not statistically significant (p>0.05).The containing melanin granules cell count of2%,4%,6%,8%of concentration of compound psoralen liniment application group concentration were compared with the saline control group. The difference was statistically significant (p<0.05).The containing melanin granules cell count of6%,8%of concentration of compound psoralen liniment application group were compared with2%,4%of one. The difference was statistically significant (p<0.05).The containing melanin granules cell count of6%of concentration of compound psoralen liniment application group were compared with8%of one.The difference was not statistically significant (p>0.05).The melanin content index(MCI)2%,4%,6%,8%of concentration of compound psoralen liniment application group concentration were compared with the saline control group. The difference was statistically significant (p<0.05).MCI of6%,8%of concentration of compound psoralen liniment application group were compared with2%,4%of one.The difference was statistically significant (p<0.05).MCI of6%of concentration of compound psoralen liniment application group were compared with8%of one. The difference was not statistically significant (p>0.05).Tyrosinase levels2%,4%,6%,8%of concentration of compound psoralen liniment application group concentration were compared with B group. The difference was statistically significant (p< 0.05).Tyrosinase levels of6%,8%of concentration of compound psoralen liniment application group were compared with2%,4%of one. The difference was statistically significant (p<0.05).Tyrosinase levels of6%of concentration of compound psoralen liniment application group were compared with8%of one. The difference was not statistically significant (p>0.05).2Clinical observation, the marked improvement rate in the experimental group was73.33%,and the marked improvement rate in the control group was36.67%.The difference was statistically significant (p<0.05). Tyrosinase levels in the experimental group were compared with that in the control group. The difference was statistically significant (p<0.05).Conclusion:Animal experiment:oral sanbai granule and topical different concentrations of compound psoralen liniment both can deepen the color of the skin of guinea pigs, improve the morphology and function of melanocytes, increase containing melanin granules cell count and MCI, and significantly increased the content of tyrosinase in the guinea pig blood. This may be one of the mechanisms of the clinical treatment of vitiligo and other hypopigmented skin.2Clinical observation:oral sanbai granule and topical6%of concentrations of compound psoralen liniment was safe and effective on vitiligo.3This treatment may play a therapeutic role by improving the level of tyrosinase concentration in the blood.
