Objeetive:To study the effects and mechanisms on Bazhen Tripe Diet of the rats on promoting gastric ulcer healing by establishing experimental chronic ulcer models of spleen deficient type.This study can provide scientific theories and experiment evidences for the clinical use of Bazhen Tripe Diet to treat the diseases of gastrie ulcer.Methods:We selected68SD rats as the study object, were randomly divided into the normal group which had18rats and the spleen deficient group which had50rats.We used the method of rhubarb combination diet dishonor to replicate spleen deficient model. After9days, select10rats from each group to test the spleen deficient model. Observe the general condition of the rats:the diet,hair,stool,activity etc; weight;swimming time;detect the immune functions:thymus and spleen index, the contents of IL-2, IgA, IgM and IgG. Divide the other rats of the spleen deficient model into five groups:model group,Bazhen Tripe Diet in low dose group, Bazhen Tripe Diet in middle dose group, Bazhen Tripe Diet in high dose group and rabeprazole group. The rats of five groups except normal group were duplicated the gastric ulcer model of spleen deficient by using acetic acid that was produeed by okabe.Give drugs after three days. Feeding after28days, we got the blood, gastric juice and the tissue. Compare the improvement of spleen deficient index, the ulcer areas, observed the tissue by eye and macroscopic to evaluate healing condition.We detected the acidity of gastric fluid and the activity of pepsin,collected blood in order to detect MDA、SOD、GSH-Px、NO、GAS、 MTL,got stomach tissue for PGE2、EGF、VEGFmRNA to discuss the possible mechanism.Results:1.Animal model:The model was duplicated by using rhubarb and dishonor diet and acetic acid that was produeed by okabe.Give rhubarb a few days,the rats’ general conditions becomed abnormal:weight lost, activity reduced, stool becomed thin and trickle, coat haggard, back arched, swimming time shortened, thymus and spleen index decreased, the contents of IL-2, IgA, IgM, IgG reduced.But after the drug treatment the above phenomenons had improved, each index had rebounded which verified that the spleen deficient model was made successfully. The stomach which was painted by acetic acid becomed pale, around the position the stomach was edema and congestive. The method of acetic acid cauterization serosa to ulcer which was used in this experiment.ulcers’performance was very similar to human ulcer through naked eye and microscopic observation.This modeling method with high success ratio was simple and easy to operate:it is now generally accepted and widely used to observe the curative effect of drugs.2.The naked eye and microscopic observationGastric mucosa appeared impairment, the depth reaching muscularis. We found that ulcer of rats in drug groups healed well compared with that of the model group,and ulcer area of the high dose group was smaller than that of other groups. We found that mucosal damagement of the model group was more serious,glands arranged disorder,there were a large number of inflammatory cells and inflammatory exudates.Each treatment group all had different degrees of healing,ulcer area was smaller than the model group, there was a small amount of inflammatory cells.3. The impact on the ulcer areaCompared with model group, ulcer area of the Bazhen Tripe Diet groups and Rabeprazole azole group decreased significantly (P<0.05or P<0.01).Bazhen Tripe Diet in high dose group was better than that of rabeprazole group (P<0.05).4. Effects on the dose and acidity of gastric juice and pepsin activityThe determination of the acidity of gastric juice had showed that there was very remarkable difference between model group and normal group(P<0.01). Compared with model group, through the acidity of gastric juice decreased in the low group, there was no difference (P>0.05); the acidity of gastric juice in the middle and high dose group and rabeprazole group significantly decreased (P<0.05or P<0.01). There was no significant difference between rabeprazole group and the high dose group(P>0.05).The determination of the dose of gastric juice and the content of pepsin activity had showed that there was very remarkable difference between model group and normal group(P<0.01). Compared with model group, through the dose of gastric juice and the content of pepsin decreased in the low group, there was no difference (P>0.05); the dose of gastric juice and the content of pepsin in the middle and high dose group and rabeprazole group significantly decreased (P<0.05or P<0.01). There was no significant difference between rabeprazole group and the high dose group(P>0.05). 5. Effects on the content of MDA, SOD and GSH-PxThe determination of the content of SOD had showed that there was very remarkable difference between model group and normal group.(P<0.01). Compared with model group, the content of SOD increased in the healing groups(P<0.05or P<0.01). Compared with rabeprazole group, the content of SOD in high dose group increased (P<0.05).The determination of the content of MDA had showed that there was very remarkable difference between model group and normal group(P<0.01). Compared with model group, the content of MDA decreased in the healing groups(P<0.05or P<0.01). There was no significant difference between the high dose groups and rabeprazole group (P>0.05).The determination of the content of GSH-Px had showed that there was very remarkable difference between model group and normal group.(P<0.01). Compared with model group, the eontent of GSH-Px increased in the healing groups (P<0.05or P<0.01). The content of GSH-Px in the high dose group inereased obviously than the rabeprazole groups (P<0.05).6. Effects on the content of NOThe determination of the content of NO had showed that there was very remarkable difference between model group and normal group(P<0.01). Compared with model group, the eontent of NO increased in the middle and high dose group and rabeprazole group(P<0.05or P<0.05). There was significant difference between the high dose group and rabeprazole group (P<0.05).7. Effects on the content of GAS and MTLThe determination of the content of GAS had showed that there was very remarkable difference between model group and normal group(P<0.01). Compared with model group, the content of GAS decreased in healing groups(P<0.05or P<0.01).There was significant difference between the high dose group and rabeprazole group (P<0.05).The determination of the content of MTL had showed that there was very remarkable difference between model group and normal group.(P<0.01). There was no significant difference between healing groups and the model group (P>0.05). But according to data there was a tendency to reduce the level of MTL8. Effects on the content of PGE2and EGFThe determination of the content of PGE2had showed that there was very remarkable difference between model group and normal group(P<0.01). Compared with model group, the content of PGE2inereased in the healing groups(P<0.05or P<0.01). There was significant difference between the high dose group and rabeprazole group (P<0.05).The determination of the content of EGF had showed that there was very remarkable difference between model group and normal group(P<0.01). Compared with model group, the content of EGF inereased in the middle and high dose group and rabeprazole group(P<0.05or P<0.01). There was remarkable difference between the high dose group and the rabeprazole group(P<0.01).9. Effect on the expression of VEGFmRNAThe determination of the content of VEGFmRNA had showed that there was very remarkable difference between model group and normal group(2-△△CT<0.5). Compared with model group, the content of VEGFmRNA inereased in the middle and high dose group and rabeprazole groups(2-△△CT>2). There was no remarkable difference between the low dose group and model group.(0.5<2-△△CT<2).Conclusion:Bazhen Tripe Diet played a promoting role on the healing gastric process which was dependent on dose.The Effeetive Mechanism might be related to gastric ulcer relieving the mucosa injury(decrease the acidity of gastric juice and pepsin activity and the content of MDA、GAS), enhancing the defence ability of gastric mucosa elevating the quality of ulcer healing(increase the content of SOD、GSH-Px、EGF、NO、PGE2、VEGFmRNA and the immune function).
