Peptic ulcer is a common and multi-occurred disease, there are about 10 percent people trouble this disease in their whole lives. Gastric ulcer is a peptic ulcer occurred from cardia to pylora. The mechanism of the occurrence and development of gastric ulcer is very complex and is not completely clarified. Generally gastric ulcer occurs as a result of the imbalance between aggressive and defensive factors affecting the gastroduodenal mucosa, the impairment of defensive factors is the main reason. Recently, owing to the extensive application of H2 recipient blocker and proton pump inhibitor, the short-term ulcer healing rate has been enhanced, but the side-effect of these drugs and relapse after drug withdrawal are problems to resolve now. Traditional Chinese Medicine(TCM) exerts the predominance on improving the gastric mucosal defensive function, increasing the quality of ulcer healing and reducing ulcer relapse. According to the TCM theory, gastric ulcer belongs to stomach ache category. Danshen Decoction was a famous medical prescription consisting of salvia miltiorrhiza, santalum album L and amomum villosum. It has an effect of promoting blood circulation to remove obstruction in the channels with a long history of therapeutic uses. Many clinical Studies have shown that Danshen Decoction has good preventive and therapeutic effects on gastric ulcer, but there has no experimental research reported by now. Our study aims to afford the theory basis for its clinic extend and application.This experiment made the gastric ulcer models by water-restraint stress in mice, acetic acid cauterization and pyloric ligation in rats. The animals were administered with different doses of Danshen Decoction respectively, then the effects of Danshen Decoction on experimental gastric ulcers were detected, and the total gastric acidity, the activity of pepsin in pyloric ligation gastric ulcer were determined. The results indicated that Danshen Decoction could reduce the acute gastric mucosal lesion, accelerate the healing of ulcer induced by acetic acid dose-dependently (This effect was of the best significance.), and also inhibit the formation of gastric ulcer induced by pylorus ligature ,while having no influence on the secretion of gastric acid and the activity of pepsin.The acetic acid-induced gastric ulcer in rats is resemble to chronic gastric ulcer in human. It is often used to investigate the effect of drugs on chronic ulcer healing. So we established acetic acid-induced gastric ulcer in rats again to explore the relative mechanisms of Danshen Decoction accelerating ulcer healing. We observed the pathology of ulcer tissues by HE staining, measured the serum nitric oxide and the plasma prostaglandin E2 by nitrate reductase method and radioimmunoassay respectively, determined the gastric wall binding mucus with AB-PAS staining, and detected ulcer margin mucosal cell apoptosis and the protein expression of epidermal growth factor receptor, basic fibroblast growth factor, Bcl-2 by TUNEL technique and SABC immunohistochemical method. The results indicated that Danshen Decoction could alleviate the lesion induced by acetic acid, accelerate regeneration of epithelial structures and re-establishment of the underlying connective tissue. Both the content of serum NO and the amount of gastric wall binding mucus in Danshen Decoction groups were higher significantly than those in the model group(P<0.01), and the level of plasma PGE2 in 3.5, 7.0g/kg groups was also higher obviously than that in the model group(P<0.05). Which all contributed to improving the gastric mucosal defensive function. Besides, Danshen Decoction could increase the protein expression of EGFR in the mucosal epithelial cells of the ulcer margin significantly(P<0.05, P<0.01), consequently stimulate the cell proliferation, differentiation and migration. In the normal group, the protein expression of bFGF was lower, while that in the model group was high obviously (P<0.01).
