Objective: To investigate the expressions and clinical significance of DJ-1genemRNA and its protein in endometrial carcinomas, paraneoplastic tissues and normalendometrial tissues.Methods: Reverse transcription polymerase chain reaction (RT-PCR), Westernblot and immunohistochemical Elivision methods are used to detect the expressionsof DJ-1gene mRNA and its protein in tumor tissues,para-tumor tissues of100casesof endometrial carcinomas and in normal endometrial tissues of30cases, thenanalyze the relationship between DJ-1gene mRNA and clinical pathologicalcharacteristics of endometrial carcinoma specimens, such as tumor histological type,differentiation, depth of invasion, lymph node metastasis, FIGO stage, serum CA125level and estrogen receptor(ER).Results: The levels of DJ-1mRNA and its protein expressions in endometrialcarcinomas are significantly higher than in para-tumor tissues and normal endometrialtissues, but the expressions between paraneoplastic tissues and normal endometrialtissues have no significant difference. There are no significantly relationshipsbetween DJ-1mRNA expression and histological type, FIGO stage, blood CA125level of endometrial carcinomas. DJ-1mRNA expressions of poorly-differentiatedand moderately-differentiated endometrial carcinomas are significantly higher thanthat of well-differentiated endometrial carcinomas, deep myometrial invasion inendometrial carcinomas DJ-1mRNA levels are higher than limited to the mucosa andsuperficial myometrial invasion endometrial carcinomas, DJ-1mRNA in endometrialcarcinoma patients with lymph node metastasis are higher than those without lymphnode metastasis, ER-negative is higher than ER-positive.Conclusions: DJ-1mRNA and its protein in endometrial carcinomas are highexpressions,The expressions of DJ-1mRNA in moderately-poorly differentiated,deep myometrial invasion, lymph node metastasis and ER-negative of endometrialcarcinoma patients are significantly increased, it may be involved in the regulations ofproliferation and apoptosis, invasion and metastasis in endometrial carcinomas,suggesting that DJ-1would be identified as an indicator of the malignant biological behavior of endometrial carcinomas, it will possibly to be a new target for moleculartherapy of endometrial carcers.
