| Objective:To investigate the expression and clinical significance of MTDH gene mRNAand its protein in endometrial carcinoma tissues and normal endometrial tissues,providing a new direction for the diagnosis of endometrial cancer.Methods:The expression of MTDH gene mRNA and its protein in83cases of endometrialcarcinoma tissues and30cases of normal endometrial tissues were detected byReverse transcription polymerase chain reaction(RT-PCR)and Western blot methods.Then,we analyzed the relationship between the expression of MTDH and clinicalpathological characteristics of endometrial carcinoma, such as tumor histological type,differentiation, depth of invasion, FIGO stage, lymph node metastasis, serum CA125,estrogen receptor(ER)and progestogen receptor (PR).Results:1. The expression levels of MTDH mRNA in endometrial carcinoma tissues aresignificantly higher than in normal endometrial tissues(P<0.001).2. The expression levels of MTDH protein in endometrial carcinoma tissues aresignificantly higher than in normal endometrial tissues(P<0.01).3.The over expression of MTDH protein was positively correlated withhistopathological grade, the depth of myometrial invasion, lymph node metastasis andFIGO stage, but not with age, tumor histological type, serum CA125, ER or PR.Conclusions:The overexpression of MTDH may be involved in the proliferation, invasion,metastasis and prognostic of endometrial carcinoma, suggesting that MTDH would beidentified as an indicator of the malignant biological behavior of endometrialcarcinoma and have positive effect on its treatment and the assessment prognosis. |
