Background Endometrial carcinoma is one of the common malignant tumors in women,,which accounts for 20%30%[1]of female genital malignant tumors.Its incidence rate is increasing in many countries,including China,which seriously reduces the quality of life and threatens the life and health of women.With the development and progress of society,people's requirement for quality of life has gradually become one of the focuses of cancer research.Endometrial carcinoma is a complicated process step by step,generally maintained in long-term estrogen stimulation without antagonistic hormone called progesterone resistance caused by endometrial hyperplasia?simple hyperplasia,complex hyperplasia,atypical hyperplasia?,on the basis of a cancerous form,this process may be involved in a variety of factors such as cancer genes,tumor suppressor genes,mismatch repair genes and cytokines.However,the exact pathogenesis of endometrial cancer is still not very clear,so it has become a hot spot of recent research.Therefore,it is necessary for us to further explore the mechanism of the occurrence and development of endometrial cancer,so as to achieve early diagnosis and early treatment,so as to reduce mortality,delay life and improve the quality of life of women.Objective To investigate the effect of adiponectin on MAPK/ERK signaling pathway in endometrial carcinoma cell line HEC-1A and Ishikawa,and further investigate the effect of adiponectin on invasion of endometrial cancer cells,so as to provide new ideas for adiponectin in the diagnosis and treatment of endometrial carcinoma.Methods Choosing 2 different degree of differentiation of endometrial carcinoma cell lines HEC-1A?moderately differentiated?and Ishikawa?IK,well-differentiated?as the experimental object,by Transwell assay and scratch experiment determination of adiponectin concentration on invasion and metastasis of endometrial cancer cells?effective concentration?;The effective concentration of adiponectin,Western Blot detection MAPK,the activation of the ERK signaling pathway and the relationship between cell invasion metastasis.All data are analyzed with statistical software SPSS 19.Results1.Effect of adiponectin on invasive ability of endometrial carcinoma cells1.1 Count the cells that pass through the membrane,and calculate the inhibition rate.With concentration gradient were 0-5-10-20?g/ml adiponectin tumor cells 24h by Transwell assay was used to observe that adiponectin concentration was greater than5?g/ml,can inhibit tumor cell invasion,the inhibition rate is 22.67%-77.35%,when the concentration increased to 10?g/ml,significantly inhibited the invasion inhibition rate with.Adiponectin concentration increases,each experimental group compared with the control group,the difference was statistically significant?P<0.01?.1.2 Count the number of cells crossing the standard distance,and observe the movement ability of the tumor cells to determine the ability of invasion.With concentration gradient were 0-5-10-20?g/ml adiponectin tumor cells treated with 24h,observed by scratch test method,the adiponectin concentration was greater than 5?g/ml,the experimental group compared with the control group over the standard distance of the cell number of experimental group decreased,when the concentration increased to10?g/ml.Significant inhibitory effect,the difference was significant?P<0.01?.The results show that the concentration of adiponectin in more than 10?/ml,has obvious inhibitory effect on the movement ability of tumor cells.2.adiponectin activates MAPK and ERK signaling pathway in HEC-1A and Ishikawa cellsDifferent concentrations of adiponectin respectively in HEC-1A and Ishikawa cell lines,Western Blot results showed that MAPK and ERK protein in the corresponding protein 2 in endometrial carcinoma cell line?cyclin D1 and cyclin E2?the relative expression levels were decreased,and the experimental group and the control group showed no significant differences?P<0.01?.The results suggest that the MAPK and ERK signaling pathway in endometrial carcinoma cells can be inhibited by adiponectin to achieve the proliferation of antitumor cells.ConclusionIn the effective concentration,adiponectin can further inhibit MAPK/ERK signaling pathway,which can inhibit endometrial cancer cell invasion and metastasis,it is no doubt that the study provides a new way of thinking and bring the dream of hope for the older age,who is in poor physical condition and can not withstand the surgery and chemotherapy toxicity in patients with clinical treatment.
