Objective:Bronchial asthma is a chronic inflammatory disorder of the airway. It plays an important role in the pathogenesis of bronchial asthma for instance airway inflammation, AHR, airway remodeling, the unbalance of Thl/Th2and so on. Lifei Jianpi recipe is a experiential prescription of TCM master Ni Zhuying for the treatment of childhood bronchial asthma remission which has many functions including invigorating the spleen, tonifying qi and propelling lung-Qi. The method by OVA sensitized excitation to build asthma animal models were adopted on clinical. To observe the effect of Lifei Jianpi recipe to IgE, IL-4, IFN-γ, ICAM-1, VCAM-1, IL-5mRNA and GR-β mRNA of SPF asthma rats by ELISA and PCR. To evaluate airway inflammation, AHR, airway remodeling the unbalance of Th1/Th2of asthma rats with Lifei Jianpi recipe. To discuss the pathogenesis of bronchial asthma with treatment of Lifei Jianpi recipe. To provide reliable justification of experiment and theory for Chinese medicine treatment of childhood bronchial asthma.Methods:Fifty rats were randomly assigned into normal control group (A, blankgroup); asthmatic group(B, Asthma Model Group); asthmatic group by Jade screen powder(C, Jade screen powder group); asthmatic group by dexamethasone treated(D,DEX group);asthmatic group by Lifei Jianpi recipe treated(E, treatment group). Every group has10 rats. The method by OVA sensi tized excitation to build asthma animal models were adopted in the experiment.To observe the affect of Lifei Jianpi recipe to serum IgE, ICAM-1, VCAM-1, IFN-γ, IL-4and lungt issue IL-5mRNA, GR-β m RNA of asthma rat with ELISA and RT-PCR. To observe lung pathological changes in different groups of rats.Result:1. The influence of serum IgE for asthma ratsCompared with blank group, the serum IgE of asthma Model Group was increased significantly (P<0.01). And the serum IgE in both of the treatment group and DEX group was decreased significantly compared to asthma Model Group (P<0.01) and Jade screen powder group (P<0.01). But no differences in serum IgE level was observed between the treatment group and DEX group (P=0.58).2. The influence of serum IL-4and IFN-γ for asthma ratsCompared with blank group, the serum IL-4of asthma Model Group was increased significantly and the serum IFN-γ decreased (P<0.05). But IL-4was decreased and IFN-γ increased with the treatment group and DEX group compared to asthma Model Group(P<0.05). It was found no significant difference in the IL-4and IFN-y expression between the treatment group and DEX group (P=0.63, P=0.10). Compared with Jade screen powder group, the treatment group had lower level of IL-4and higher level of IFN-γ. And the difference was significant (P<0.05).3. The influence of serum ICAM-1and VCAM-1for asthma ratsCompared with blank group, the lever of serum ICAM-1and VCAM-1was increased significantly in asthma Model Group (P<0.05). And compared to asthma Model Group, the lever of ICAM-1and VCAM-1fell significantly among the treatment group, DEX group and Jade screen powder group (P<0.05). But there was no difference between the treatment group and DEX group(P=0.28, P=0.87). Compared with Jade screen powder group, the treatment group had lower level and the difference was significant (P<0.05).4. The influence of lungtissue IL-5mRNA for asthma ratsCompared to blank group, the lung tissue IL-5mRNA of asthma Model Group increased significantly (P<0.05). Compared with the asthma Model Group, the level of the lungtissue IL-5mRNA of treatment group and DEX group were decreased significantly (P<0.05). The IL-5mRNA of the treatment group had lower expression in the lungtissue compared to DEX group (P<0.05). It was no significant differences between the treatment group and Jade screen powder group (P=0.46).5. The influence of lungtissue GR-β mRNA for asthma ratsCompared to blank group, the lungtissue GR-β mRNA of asthma Model Group fell Sign if icantly (P<0.05). Compared with asthma Model Group, the lungtissue GR-β mRNA of the treatment group was increased significantly (P<0.05). And compared with Jade screen powder group and DEX group, the lungtissue GR-β mRNA of the treatment group had greater expression (P<0.05).6. The influence of lung Pathological changes for asthma ratsIn the treatment group which was treated with Lifei Jianpi recipe, the trachea tube wall gradually was recovered, less inflammation, and the edema of tissue was obviously disappeared. Meanwhile there was less inflammatory exudate inside lumen.Conclusions:1. It is a feasible method to build asthma rats model with OVA sensitized excitation. 2. We could find that Lifei Jianpi recipe make Thl/Th2scale of asthma rat verge to balance, airway inflammation of asthma rat remitted, AHR and airway remodelling restrained, and aggregation of Eosnophils restrained. And it could promote Glucocorticoid Receptor to combine with glucocorticoid.
