Objective To investigate whether the human placenta expresses immunereactive EPO and EPOR.Methods The localization and immunoreactivity of EPO and EPOR are detected by SP immunohistochemistry and Western blotting. Positive comparison film served as a positive control and sections with PBS instead of Primary antibody as a negative control.Results1. Villous and extravillous cytotrophoblast cells, as well as syncytiotrophoblast at all gestational stages, expressed immunoreactive EPO and EPO-R identified by immunohistochemistry.2. placental tissues of various gestational ages, also expressed immunoreactive EPO and EPO-R by Western blot.Conclusions1. This study demonstrated that fetoplacental tissues expressed EPO and EPOR at various gestational ages, further, as the gestational age developing, the immunoreactive EPO and EPOR more intense.2. By analogy to the action of the colony-stimulating factors on trophoblast cells, and of these hormones as well as EPO on bone marrow precursors, we postulate that the placenta is translationally active in producing the EPO hormone and placental EPO may be a significant and unique pregnancy factor supporting fetal growth and development.
