Objective:To investigate the mechanism of yi qi ding xuan yin based on EPO/EPOR channel against cerebral ischemia in rats.Methods:The cerebral ischemia model of rats was established by the middle cerebral artery occlusion method(based on the modified Longa method).A total of 120 healthy male SD rats were randomly divided into sham operation group,model control group,butylphthalide group,and yi qi ding xuan yin group.They were randomly divided into 1d group,3d group,7d group and 14 d group according to different intervention time,with at least 5animals in each group.The neurological function scores of rats were observed at each time point.HE staining was used to observe the pathological changes of the ischemic brain tissue of rats with cerebral ischemia injury.Apoptotic cells were detected by TUNEL assay,and the expressions of brain tissue proteins EPO and EPOR were detected by immunohistochemistry.Results: Compared with the model control group,the scores of neurological deficits in the butylphthalide group and the yi qi ding xuan yin group were significantly lower on 3d,7d and 14d(P<0.05).Compared with the sham operation group,the apoptosis rate of the model control group was increased(P<0.01).Compared with the model control group,the apoptosis rate of the butylphthalide group and the yi qi ding xuan yin group was significantly decreased on 3d,7d and 14d(P<0.05).Compared with the sham operation group,the number of EPO and EPOR positive cells was significantly increased in the model control group on 3d,7d and 14d(P<0.05),and the number of EPO and EPOR positive cells was significantlyincreased in the butylphthalide group and the yi qi ding xuan yin group on3 d,7d and 14d(P<0.05).Conclusion: The effects of modified yi qi ding xuan yin and butylphthalide on the nerve function and local neuron morphology after cerebral ischemia in rats were improved,and the effect was more significant in the 14 d group,which may be related to the production of EPO and EPOR proteins.
