Objective In this study, according to established the iron deficiency Wister rats model, and then detected the hematological parameters, serum erythropoietin, growth hormone(GH), insulin-like growth factor-1(IGF-1) levels, the expression of EPOR in rats hypothalamus, pituitary and adrenal, and measured the rats’ weight and body length. Preliminarily investgate the rats iron deficiency impact on the hypothalamus,pituitary, adrenal and GH-IGF-1 axis.Methods 40 Wistar rats were randomly divided into two groups after one week adaptive feeding. The experimental group were fed with low-iron diet, while the control group were fed with normal diet.Detected the rats hematological indices by inner canthus vein blood every week, and measured body length and weight. After 4-week-low iron diet intervention, detected the serum concentration of EPO, GH and IGF-1,and detected the expression of EPOR in hypothalamus, pituitary, adrenal tissues by immunohistochemistry.Result(1) After 4 weeks, the concentrations of hemoglobin(122.64 ± 9.55 g/L) and ferritin(79.77 ±24.99umol/L) decreased in the experimental group(P<0.05),so the models is good.(2) Two groups of rats’ weight were increased, but a smaller increase happened in the experimental group; The serum concentration of GH, IGF-1 in experimental group was obviously lower than the control group, the difference was statistically significant(P<0.05).(3) Immunohistochemical results showed that in the control group the expression of EPOR in hypothalamus is very low, most got 0 point and 1 point while high expression only had 12.5%;The expression of EPOR in the experimental group also had a low level, but 1point and 2 points rates was higher; the experimental group higher expression rate were obviously higher than the control group(P<0.05).During the study, we found that the expression in neurohypophysis was more obvious than anterior lobe. Meanwhile, EPOR can expressed both in the adrenal cortex and medulla,and the experimental group higher expression rate were obviously higher than the control group(P<0.05).Conclusion(1) Iron deficiency can cause the rats serum concentrations of GH, IGF-1 reduce, then inhibit the GH-IGF-1 axis result in affecting their growth and development.(2) Iron deficiency can also cause the expression of EPOR in rats hypothalamus, pituitary, adrenal increased to accommodate tissues and organs ischemic and hypoxia, in order to protect the tissues and organs.(3) EPOR can protect the iron deficiency rats hypothalamic-pituitary-adrenal axis.
