Objective:To investigate the expression of ADM、EPOR in human primary hepatocellularcarcinoma and its relationship with the tumor pathological characteristics. And to detectthe relationship among the expression of ADM and EPOR with tumor size, tumormetastasis, pathologic types and other biocharacteristics of the mutual relations. Toinvestigate the role of ADM、EPOR in hepatocellular carcinoma angiogenesis and itspotential clinical applications for cancer therapy.Method:Fresh tumor and normal liver tissues were obtained from30hepatocellular carcinomapatients after hepatectomy.10hepaticcirrhotics and10normal liver cantrols were studied.Expression levels of ADMmRNA and EPORmRNA were quantitatively analyzed byreal-time PCR. The microvessels were immunohistochemically labeled with CD34toassess microvessal density. The relationship between the ADM、erythropoietin receptor andpathological features as well as microvessel density(MVD) was evaluatsd.Results:1. The expression of ADMmRNA and EPORmRNA in the cancer tissues were muchhigher than that in the non-cancer tissues(P<0.05), while tissues levels inhepaticcirrhotics group and normal liver group have no significant difference (P>0.05).2. Over expression of ADMmRNA and EPORmRNA in hepatocellular carcinoma tissueswas related to the invasion of tumor envelop,pathological differentiation and tumormetastasis(P<0.05). The expression of ADMmRNA and EPORmRNA has norelationship with gender、age、tumor size、AFP level and hepaticcirrhotics (P>0.05). 3. MVD in liver cancer tissues was associated with tumor size(t=2.87,P<0.05),theinvasion of tumor envelop(t=3.94,P<0.05),tumor metastasis(t=4.13,P<0.05)andpathological differentiation (t=5.25,P<0.05).4. Correlation analysis to demonstrate that ADM and EPOR, ADM and MVD,EPOR andMVD have significantly correlation, the coefficient correlation are respectively0.74、0.68、0.71(P<0.01).Conclusions:1. Our study shows that the expression of ADM and EPOR are significantly higher inhepatocellular carcinoma group, the high expression of them were related to theinvasion of tumor envelop, pathological differentiation and tumor metastasis.2. Expression of MVD in hepatocellular carcinoma group was associated with tumor size,invasion of tumor envelop, tumor metastasis and pathological differentiation.3. ADM and EPOR may serve as new angiogenesis regulatory factors involved inangiogenesis of hepatocellular carcinoma, to interfere with ADM、EPOR and theirreceptor pathway are expected to be a potential target for anti-angiogenesis therapy ofhepatocellular carcinoma.
