At present, the most prevalent kind of biochip is a tiny module, fabricated withmicro-machining techniques and molecular biological techniques to have somedetection and analysis ability of molecular biology It can be used to accomplish aseries of procedures of biological sample- separation. preparation, biochemicalreaction, detection of product and so on. It is divided into two kinds of biochip-probe array chip and integrated capillary electrophoresis chip.Integrated capillary electrophoresis chips (also called Lab-on-a-chip chip) ismultifunctional, fast, high-efficiency and low-power-consumption laboratorytechniques, which make use of routine capillary electrophoresis techniques onseveral square centimeter size chip, and which is a system consisting of sampleinjector, reactor, separation capillary and detector. It features miniaturization,integration and automation of separation and determination. Thus it is the advancearea of modern analytical chemistry.Capillary electrophoresis chip is key functional component of Lab-on-a-Chip chip,on which the aspect of groove is less than one hundred micron. In this dissertation,by using MEMS techniques the chip has 100×μm cross-section channelsmicrofabricated in a 2×3 centimeter glass. with thermal bonding method, the chipwith cross column geometry and the chip with serpentine column geometry arerealized.Detector is an important component of integrated capillary electrophoresis chips;its sensitivity and precision have direct influence on the performance of chips. In thisdissertation, by using photomultiplier tube and photoconductive audion respectively,the software and hardware of detector are realized. Photoconductive audion is apotentially integrated component, thus this dissertation discuss its performance.The fundamental theory of manipulation of sample and electrophoresis of sampleon the chip is electrodynamics. In this dissertation, by using electrodynamics, someelementary experiments are carried out. Amino acids are derived by the use of 2,4-dinitrofluorobenzen (DNFB), its products are used to realize manipulation of sampleon the chip, and the affecting factors of electrophoresis are also discussed.
