Objective:To adopt ibotenic acid (IBO) and the AmyloidβProtein Fragment 1-42 (Aβ1-42) of the mixed inject to sea horse area establish AD rat models,based on pathogenic mechanism of kidney-Qi deficiency and phlegm stasis in Alzheimer's disease,To observe the AD rat model of brain behavior influence and immunohistochemistry was used to detect growth factor in rat sea horse TGF-ɑ, IGF-Ⅱlevels in, to explore Buyuan Congnao Decoction(BCD) in the treatment of senile dementia and the possible mechanism.Methods:75 healthy SD male rats were randomly divided into 5 groups:Normal group,Control group, Model group, Chinese traditional medicine group (BCD), western medicine group (piracetam group). In addition to Normal group and Control group, the remaining three groups of rats were injected into the sea horse of Aβ1-42 and the IBO mixture made of AD rat model,sea horse of Control group injected normal saline. After two weeks of modeling, Chinese traditional medicine in rats given a daily Buyuan Congnao Decoction(BCD) (11.1g/Kg), Western medicine in rats given piracetam (324mg/Kg), except the treatment group, the rest of the group were given normal saline gavage, 28 consecutive days once a day, Detected by using Morris water maze learning and memory abilities in rats. Observed pathological morphology of hippocampus by the light microscope. Then detected by immunohistochemistry in neurons growth factor TGF-ɑ, IGF-Ⅱlevel expression.Results: Ethology: Compared with normal group, latency period of escape in model group prolonged obviously (P﹤0.01), frequency of finding the platform decreased(P﹤0.01),learning and memory in rats significantly decreased, indicating a successful modeling. After take Buyuan Congnao Decoction(BCD) and piracetam, Chinese traditional medicine and western medicine group compared with model group, latency period of escape shortened(P﹤0.01), frequency of finding the platform increased(P﹤0.01), Chinese traditional medicine and western medicine group rats significantly improved learning and memory capability.Morphological changes under light microscope: Number of neuron of model group was less than that of normal group, neurons of Chinese traditional medicine and western medicine group repair more obvious, nuclear pyknosis and vacuolar degeneration of certain improvements, neurofibrillary tangles and amyloid deposits decreased neuron number than the model group, and the cell structure is also arranged complete than the model group.Growth factor TGF-ɑ, IGF-Ⅱ: Response model group was significantly stronger than the normal group and control group, the difference was significant (P﹤0.01). Chinese medicine and western medicine group were compared with model group,TGF-ɑ, IGF-Ⅱresponse intensity decreased, the number of positive cells was significant(P﹤0.01).Conclusions:1.Using A beta 1-42 and IBO multi-factor model of the composite mixture made dementia model has its feasibility.2.Dementia is the basic pathogenesis of deficiency of phlegm stasis and its effective therapeutic resistance is remove stasis phlegm kidney.3.Buyuan Congnao Decoction(BCD) can significantly improve learning and memory capability in rats with Alzheimer.4. Buyuan Congnao Decoction(BCD) can significantly improve the AD pathology in hippocampal neurons.5. Buyuan Congnao Decoction(BCD) may be by regulating the brain tissue growth factor TGF-ɑ, IGF-Ⅱlevels,then affectβ-amyloid generation to achieve the treatment of AD role.6. Buyuan Congnao Decoction(BCD) can improve learning and memory in Alzheimer's ability, preventing plays an important role in the process.
